Gut Microbiota Regulates Systemic Inflammatory Response and Compensatory Anti‐Inflammatory Response Syndromes by Targeting PF4+ Macrophages in Acute Pancreatitis

open access: yesAdvanced Science, EarlyView.
ABSTRACT Acute pancreatitis (AP) begins with pancreatic local inflammation, leading to the onset of systemic inflammatory response syndrome (SIRS), followed by compensatory anti‐inflammatory response syndrome (CARS), which causes immune paralysis and higher mortality rate.

CDK4/6 Inhibition Induces CD8+ T Cell Antitumor Immunity via MIF‐Induced Functional Orchestration of Tumor‐Associated Macrophages

open access: yesAdvanced Science, EarlyView.
CDK4/6 inhibition promotes CD8+ T cell expansion through tumor‐macrophage crosstalk by activating HIF‐1α and enhancing MIF‐CD44/CD74 signaling. This reprograms TAMs to boost MHC‐I antigen presentation, and CDK4/6 inhibitor‐trained M1 TAM supernatant therapy synergizes with low‐dose PD‐1 blockade to restore antitumor immunity.

Enzymatic Activity of HPGD in Treg Cells Suppresses Tconv Cells to Maintain Adipose Tissue Homeostasis and Prevent Metabolic Dysfunction.

open access: yesImmunity, 2019
Regulatory T cells (Treg cells) are important for preventing autoimmunity and maintaining tissue homeostasis, but whether Treg cells can adopt tissue- or immune-context-specific suppressive mechanisms is unclear.

rWTC‐MBTA Vaccine, Alone and Enhanced with Anti‐PD1, Elicits Immune Responses against CNS and Peripheral B‐Cell Lymphoma

open access: yesAdvanced Science, EarlyView.
An autologous whole‐tumor‐cell vaccine (rWTC‐MBTA) is evaluated in murine CNS lymphoma. Subcutaneous vaccination activates dendritic cells, broadens T‐cell priming, and drives lymphocyte trafficking to brain tumors, producing durable tumor control. Longitudinal bioluminescence and adoptive‐transfer assays verify CNS engagement. Combination with anti‐PD‐

Th17 cells and Tregs: unlikely allies [PDF]

open access: yesJournal of Leukocyte Biology, 2014
ABSTRACTIdentification of CD4+Foxp3+ Tregs and Th17 modified the historical Th1–Th2 paradigm. Currently, the Th17–Tregs dichotomy provides a dominant conceptual framework for the comprehension of immunity/inflammation and tolerance/immunosuppression in an increasing number of diseases. Targeting proinflammatory Th17 cells or immunosuppressive Tregs has

Engineering Osteoimmune Responses with Functionalized Orthopedic Implants for Post‐Operative Osteosarcoma Treatment

open access: yesAdvanced Science, EarlyView.
Osteosarcoma is the most common primary bone tumor with limited treatment options and a terrible prognosis. This review provides a comprehensive summary of the recent development of osteoimmunomodulatory implants for post‐operative osteosarcoma treatment, of which the potential utility in evoking durable anti‐osteosarcoma immunity and accelerating bone

Throwing Treg cells off balance [PDF]

open access: yesNature Reviews Cancer, 2019
Di Pilato et al. selectively modulated the function of regulatory T (Treg) cells in tumours by targeting the CARMA1–BCL10–MALT1 (CBM) signalosome complex in only a fraction of Treg cells and showed that this was sufficient to impede tumour growth and prime the tumour microenvironment for immune checkpoint blockade.

Targeting Lactate and Lactylation in Cancer Metabolism and Immunotherapy

open access: yesAdvanced Science, EarlyView.
Lactate, once deemed a metabolic waste, emerges as a central regulator of cancer progression. This review elucidates how lactate and its epigenetic derivative, protein lactylation, orchestrate tumor metabolism, immune suppression, and therapeutic resistance.

Understanding and Overcoming Antibody‐Drug Conjugate Resistance: Biological Mechanisms and Emerging Analytical Frameworks in Breast Cancer

open access: yesAdvanced Science, EarlyView.
Antibody–drug conjugates (ADCs) transform breast cancer therapy, yet resistance limits their durability. Emerging evidence reveals that ADC failure is not solely tumor‐intrinsic but shaped by dynamic tumor–microenvironment interactions that alter drug delivery, processing, and response.

Generation of RORγt+ Antigen-Specific T Regulatory 17 Cells from Foxp3+ Precursors in Autoimmunity

open access: yesCell Reports, 2017
Th17 cells are potent mediators in autoimmune diseases, and RORγt is required for their development. Recent studies have shown that RORγt+ Treg cells in the gut regulate intestinal inflammation by inhibiting effector T cell function. In the current study,
