Toward Practical Embryo Genomic Selection in Cattle: An Optimized Workflow Enabling High-Accuracy Breeding Value Prediction Without Compromising Embryo Survival. [PDF]

open access: yesFASEB J
A diameter‐guided workflow was established to define the appropriate trophectoderm (TE) biopsy area window for bovine blastocysts, with the aim of balancing embryo viability and genotyping reliability. After TE biopsy, sampled cells were subjected to DNA amplification and genomic evaluation, while the corresponding biopsied embryos were transferred to ...

Trophectoderm Lineage Determination in Cattle [PDF]

open access: yesDevelopmental Cell, 2011
The trophectoderm (TE) and inner cell mass (ICM) are committed and marked by reciprocal expression of Cdx2 and Oct4 in mouse late blastocysts. We find that the TE is not committed at equivalent stages in cattle, and that bovine Cdx2 is required later, for TE maintenance, but does not repress Oct4 expression.

The mammalian embryo’s first agenda: making trophectoderm

open access: diamondThe International Journal of Developmental Biology, 2019
One of the bottlenecks for a successful pregnancy in mammalian species is the implantation of the early embryo into the wall of the mother’s uterus. The first cell lineage the embryo sets aside following fertilization is the trophectoderm – a specialized cell type that establishes contact with the mother and mediates embryo implantation.

Development of an Improved in vitro Model of Bovine Trophectoderm Differentiation

open access: yesFrontiers in Animal Science, 2022
The mechanisms regulating early stages of placentation and trophectoderm differentiation in the ruminant conceptus remain poorly understood. Here we present a model of trophectoderm (TE) differentiation in vitro from outgrowths of individual in vitro ...

Cytoplasmic strings between ICM and mTE are a positive predictor of clinical pregnancy and live birth outcomes: A time-lapse study

open access: yesFrontiers in Medicine, 2022
BackgroundElective single blastocyst transfer (eSBT) is considered to reduce the incidence of multiple pregnancy compared to double embryo transfer. Blastocyst selection is the key to achieving pregnancy.

Modeling human trophoblast, the placental epithelium at the maternal fetal interface. [PDF]

open access: yes, 2020
Appropriate human trophoblast lineage specification and differentiation is crucial for the establishment of normal placentation and maintenance of pregnancy. However, due to the lack of proper modeling systems, the molecular mechanisms of these processes

High-resolution transcriptional and morphogenetic profiling of cells from micropatterned human ESC gastruloid cultures

open access: yeseLife, 2020
During mammalian gastrulation, germ layers arise and are shaped into the body plan while extraembryonic layers sustain the embryo. Human embryonic stem cells, cultured with BMP4 on extracellular matrix micro-discs, reproducibly differentiate into ...

Mesenchymal Stem Cells in Embryo-Maternal Communication under Healthy Conditions or Viral Infections: Lessons from a Bovine Model

open access: yesCells, 2022
Bovine mesenchymal stem cells are a relevant cell population found in the maternal reproductive tract that exhibits the immunomodulation capacity required to prevent embryo rejection.

Essential roles of HDAC1 and 2 in lineage development and genome-wide DNA methylation during mouse preimplantation development

open access: yesEpigenetics, 2020
Epigenetic modifications, including DNA methylation and histone modifications, are reprogrammed considerably following fertilization during mammalian early embryonic development.

Trophectoderm cell failure leads to peri-implantation lethality in Trpm7-deficient mouse embryos

open access: yesCell Reports, 2021
Summary: Early embryogenesis depends on proper control of intracellular homeostasis of ions including Ca2+ and Mg2+. Deletion of the Ca2+ and Mg2+ conducting the TRPM7 channel is embryonically lethal in mice but leaves compaction, blastomere polarization,
