Elektrophysiologische Charakterisierung von TRPM3 Ionenkanälen [PDF]
Ion channels are transmembrane proteins that provide pathways for the movement of ions across cellular membranes. Ion channels play a cruical role in the regulation of fundamental life processes and are therefore found in the membranes of every cell. In order to understand the physiological function of an ion channel, knowledge about its activation and

Inhibition of Transient Receptor Potential Melastatin 3 ion channels by G-protein βγ subunits
Transient receptor potential melastatin 3 (TRPM3) channels are activated by heat, and chemical ligands such as pregnenolone sulphate (PregS) and CIM0216.

Structural basis for agonist and heat activation of nociceptor TRPM3. [PDF]
Detecting noxious heat is vital for survival, triggering protective pain responses. The TRPM3 channel is a key nociceptor and a promising therapeutic target for pain and neurological disorders. Here we show that the rabbit TRPM3 is intrinsically dynamic, with its intracellular domain (ICD) sampling both resting and activated states, but favoring the ...

Regulation of the Ion Channel TRPM3 by Phosphoinositides [PDF]
TRPM3 belongs to the Melastatin family of Transient Receptor Potential (TRP) ion channels. It is a Ca2+ permeable outwardly rectifying nonselective cation channel. TRPM3 is expressed in sensory neurons, brain, pancreas, kidney and vascular smooth muscle. In sensory dorsal root ganglion (DRG) neurons it was shown to function as a sensor for noxious heat.

TRPM3 - A Promising Target for Analgesic Treatment [PDF]
According to the American Pain Society, pain is one of the most common symptoms why patients search medical attention. A recent achievement in pain research was the identification of Transient Receptor Potential (TRP) channels as analgesic targets. The superfamily of TRP ion channels consists of 28 different members in mammalians.

Regulation of the transient receptor potential channel TRPM3 by phosphoinositides [PDF]
The transient receptor potential (TRP) channel TRPM3 is a calcium-permeable cation channel activated by heat and by the neurosteroid pregnenolone sulfate (PregS). TRPM3 is highly expressed in sensory neurons, where it plays a key role in heat sensing and inflammatory hyperalgesia, and in pancreatic β cells, where its activation enhances glucose-induced

Transient receptor potential vanilloid 1 (TRPV1) was the first noxious heat‐sensitive channel discovered. In rodents, its role is robust in the heat response of the cell body of polymodal nociceptors, but surprisingly small in that of the peripheral terminals in the skin.

G protein βγ subunits inhibit TRPM3 ion channels in sensory neurons
Transient receptor potential (TRP) ion channels in peripheral sensory neurons are functionally regulated by hydrolysis of the phosphoinositide PI(4,5)P2 and changes in the level of protein kinase mediated phosphorylation following activation of various G

Peripheral targets for neuropathic pain
Neuropathic pain represents a significant clinical challenge, with still limited pharmacological approaches to symptomatic relief. This review focuses on molecular targets implicated in neuropathic pain, particularly those involved in peripheral mechanisms. Using the IUPHAR/BPS database of biological targets, their occurrence together with ‘neuropathic
