Mucolipin co-deficiency causes accelerated endolysosomal vacuolation of enterocytes and failure-to-thrive from birth to weaning. [PDF]
During the suckling period, intestinal enterocytes are richly endowed with endosomes and lysosomes, which they presumably utilize for the uptake and intracellular digestion of milk proteins. By weaning, mature intestinal enterocytes replace those rich in

TRPML3‑mediated lysosomal Ca2+ release enhances drug sequestration and biogenesis, promoting osimertinib resistance in non‑small cell lung cancer. [PDF]
Lysosomes and lysosomal Ca2+ play crucial roles in cellular homeostasis and drug resistance. The lysosomal Ca2+ channel transient receptor potential mucolipin 3 (TRPML3; also known as mucolipin‑3 or MCOLN3) is a key regulator of autophagy and membrane trafficking; however, its role in tyrosine kinase inhibitor (TKI) resistance remains unclear.

The Ca2+ Channel TRPML3 Regulates Membrane Trafficking and Autophagy [PDF]
TRPML3 is an inward rectifying Ca2+ channel that is regulated by extracytosolic H+. Although gain‐of‐function mutation in TRPML3 causes the varitint‐waddler phenotype, the role of TRPML3 in cellular physiology is not known. In this study, we report that TRPML3 is a prominent regulator of endocytosis, membrane trafficking and autophagy.

The Concise Guide to PHARMACOLOGY 2023/24: Ion channels
The Concise Guide to PHARMACOLOGY 2023/24 is the sixth in this series of biennial publications. The Concise Guide provides concise overviews, mostly in tabular format, of the key properties of approximately 1800 drug targets, and over 6000 interactions with about 3900 ligands. There is an emphasis on selective pharmacology (where available), plus links

Abstract figure legend Amphipathic weak bases like CatSper blockers (B) diffuse across the sperm plasma membrane from a region of high concentration to one of low concentration (1). Inside the cell, these compounds are accumulated and protonated (BH+) into the acrosomal lumen (2).

This study investigates the impact of a pungent compound from ginger, [6]‐gingerol, on the immune responses of a specific human blood leukocyte cell type, the neutrophils. The study observes that [6]‐gingerol facilitates the production of reactive oxygen species and the secretion of a chemokine (CXCL8) upon subsequent stimulation with a bacterial ...

Skin melanocytes express TRPML3.
(A) Images showing sections of the back skin. τGFP expression is visible in hair follicles (HF). (B) Trpml3 levels in back skin correlate with initial synchrony of the hair follicle growth cycle. Follicle growth phases (anagen I, P9; anagen II, P27) show

Effective chiral pool synthesis of both enantiomers of the TRPML inhibitor trans‐ML‐SI3
An effective chiral pool synthesis of both enantiomers of the TRPML inhibitor trans‐ML‐SI3 was worked out on two different routes, both starting from enantiomerically pure cis‐2‐aminocyclohexanols. The absolute configuration of the more active enantiomer was elucidated. These chiral pool syntheses provide sufficient amounts of the enantiomerically pure

A TRP Family Based Signature for Prognosis Prediction in Head and Neck Squamous Cell Carcinoma
Purpose. Head and neck squamous cell carcinoma (HNSCC) is a classical type of head and neck cancers, with heterogeneous clinical outcome. This project is set out to create a robust risk signature based on TRP family genes (TFGs) for prognosis evaluation in HNSCC. Methods.
