Differential Effects of Combined ATR/WEE1 Inhibition in Cancer Cells
Inhibitors of WEE1 and ATR kinases are considered promising for cancer treatment, either as monotherapy or in combination with chemo- or radiotherapy. Here, we addressed whether simultaneous inhibition of WEE1 and ATR might be advantageous.

Cdk1-Dependent Regulation of the Mitotic Inhibitor Wee1 [PDF]
The Wee1 kinase phosphorylates and inhibits cyclin-dependent kinase 1 (Cdk1), thereby delaying entry into mitosis until appropriate conditions have been met. An understanding of the mechanisms that regulate Wee1 should provide new insight into how cells make the decision to enter mitosis.

ABSTRACT We investigated the potential of miR‐640 to downregulate CDK4/6 inhibitor resistance‐associated genes and evaluated its apoptotic, anti‐metastatic and anti‐proliferative effects, both alone and in combination with abemaciclib in breast cancer cells. miR‐640 was identified via GSE126125 dataset analysis.

YTHDF2 promotes mitotic entry and is regulated by cell cycle mediators.
The N6-methyladenosine (m6A) modification regulates mRNA stability and translation. Here, we show that transcriptomic m6A modification can be dynamic and the m6A reader protein YTH N6-methyladenosine RNA binding protein 2 (YTHDF2) promotes mRNA decay ...

ABSTRACT Toxoplasma gondii infection poses potential risks to stem cell‐based therapeutic applications; however, the effects of parasite‐derived soluble factors on mesenchymal stem cells remain unclear. Here, we investigated the impact of T. gondii‐derived soluble factors (Tg‐SFs) on human umbilical cord‐derived mesenchymal stem cells (hUC‐MSCs).

Perturbation of cytokinin and ethylene-signalling pathways explain the strong rooting phenotype exhibited by Arabidopsis expressing the Schizosaccharomyces pombe mitotic inducer, cdc25 [PDF]
Background Entry into mitosis is regulated by cyclin dependent kinases that in turn are phosphoregulated. In most eukaryotes, phosphoregulation is through WEE1 kinase and CDC25 phosphatase.

Chromosomal Instability Drives Glioblastoma Heterogeneity and Therapeutic Opportunities
ABSTRACT Glioblastoma, the most aggressive and lethal form of brain cancer, is defined by profound genomic instability, with Chromosomal Instability (CIN) playing a central role in driving tumor progression, therapy resistance, and poor prognosis. CIN is characterized by numerical and structural alterations, is driven by mechanisms such as mitotic ...

Activation domain-dependent degradation of somatic Wee1 kinase
Cell cycle progression is dependent upon coordinate regulation of kinase and proteolytic pathways. Inhibitors of cell cycle transitions are degraded to allow progression into the subsequent cell cycle phase.
