Palliative care in patients who receive whole brain radiotherapy for brain metastases in Ahmadu Bello University Teaching Hospital, Zaria [PDF]

open access: yes, 2016
Background: Brain Metastases is a devastating complication of Cancer affecting 10-50% of patients with systemic disease. It by far outnumbers primary Brain tumor in a 10:1 ratio.Aims and Objective: To determine the age distribution, gender distribution ...

Discovery and Optimization of AMT‐676, a CDH17‐Targeting ADC for the Treatment of Advanced Gastrointestinal Cancers

open access: yesAdvanced Science, EarlyView.
AMT‐676 is a novel antibody‐drug conjugate targeting CDH17, an adhesion molecule uniquely exposed on gastrointestinal tumor surfaces. Engineered with an optimized exatecan payload, it demonstrates profound tumor regression and a robust bystander effect across diverse preclinical models.

Radiotherapy Modality and Primary Tumor Type as Determinants of Outcomes in Brain Metastases: A Retrospective Study of 734 Patients

open access: yesMedicina
Background and Objectives: Radiotherapy (RT) is a standard component of palliative treatment for brain metastases. The aim of this retrospective analysis of consecutive patients treated between 2017 and 2023 is to analyze overall survival after brain ...

Targeting LDHA Palmitoylation Sensitizes GBM to Chemoradiotherapy via Suppressing Palmitic Acid Induced Glycolytic Reprogramming

open access: yesAdvanced Science, EarlyView.
Mesenchymal GSCs preferentially uptake palmitic acid, thereby promoting ZDHHC12‐mediated LDHA palmitoylation. This modification enhances LDHA enzymatic activity, drives glycolytic reprogramming, and facilitates exosomal LDHA‐mediated TAM regulation.

Acoustically Activatable Drug‐Loaded Nanodroplets for Mechanochemical Therapy in Solid Tumors

open access: yesAdvanced Science, EarlyView.
Microfluidically fabricated drug‐loaded nanodroplets are sequentially activated by dual‐frequency ultrasound through acoustic droplet vaporization followed by bubble collapse, enabling spatially controlled drug release and mechanical tumor fractionation, resulting in synergistic mechanochemotherapy and prolonged survival.

Optimizing Cancer Vaccinations Using a Physiologically‐Based Pharmacokinetic/Pharmacodynamic (PBPK/PD) Model

open access: yesAdvanced Science, EarlyView.
Where and when should a cancer vaccine be administered? A whole‐body PBPK model demonstrates that systemic immune transport critically determines vaccination efficacy. By simulating how organ‐specific immune interactions influence treatment response, the study provides a quantitative framework for optimizing cancer vaccination strategies.

AARS2‐SIRT5‐Driven DLD K445 Lactylation Promotes Cuproptosis Resistance in Glioblastoma Stem Cells

open access: yesAdvanced Science, EarlyView.
Lactylation of DLD at K445 by AARS2 suppresses PDH complex activity and reduces DLAT lipoylation, thereby conferring cuproptosis resistance in glioblastoma stem cells. SIRT5‐mediated delactylation restores PDH function and enhances cuproptosis sensitivity.

Stereotactic radiotherapy versus whole-brain radiotherapy for brain metastases

open access: yes
Moždane metastaze su najčešće dijagnosticirani maligni tumori središnjeg živčanog sustava, a uslijed napretka u dijagnostici i liječenju njihova incidencija i dalje raste.
