Xylose isomerases from Thermotoga

2001
Publisher Summary Typically present in microorganisms that grow on xylose, xylose isomerise (XI) converts xylose to xylulose, which is then phosphorylated and enters the pentose-phosphate pathway. Because it also accepts glucose as substrate, XI is used extensively to isomerize glucose to fructose in the manufacture of high fructose corn syrup (HFCS).

Reduction of PDC1 expression in S. cerevisiae with xylose isomerase on xylose medium

Bioprocess and Biosystems Engineering, 2011
Ethanol production using hemicelluloses has recently become a focus of many researchers. In order to promote D: -xylose fermentation, we cloned the bacterial xylA gene encoding for xylose isomerase with 434 amino acid residues from Agrobacterium tumefaciens, and successfully expressed it in Saccharomyces cerevisiae, a non-xylose assimilating yeast. The

Hyperexpression of Escherichia coli Xylose Isomerase

Biotechnology Progress, 1986
AbstractThe xylose isomerase (xylA) structural gene was cloned under the control of the tac promoter and expressed in a xyl+ E. coli strain. Xylose isomerase accounted for approximately 28% of the total cell protein when this tac‐xylA fusion was induced with isopropylthio β‐D‐galactopyranoside.

Simultaneous bioconversion of glucose and xylose to ethanol by Saccharomyces cerevisiae in the presence of xylose isomerase

Applied Microbiology and Biotechnology, 2000
Simultaneous isomerisation and fermentation (SIF) of xylose and simultaneous isomerisation and cofermentation (SICF) of a glucose/xylose mixture was carried out by Saccharomyces cerevisiae in the presence of xylose isomerase. The SIF of 50 g I(-1) xylose gave an ethanol concentration and metabolic yield of 7.5 g l(-1) and 0.36 g (g xylose consumed)(-1).

Streptomyces glucose/xylose isomerase has a single active site for glucose and xylose

Biochemical and Biophysical Research Communications, 1989
A kinetic method which allows one to evaluate whether an enzyme acting on two different substrates has one or two active sites was employed to study the active site of glucose isomerase which catalyses the isomerization of both glucose and xylose. The experimental data on the rates of hydrolysis of mixtures of various concentrations of glucose and ...

Overproduction of E. coli xylose isomerase

Biotechnology Letters, 1986
A promoterles DNA fragment containing theE. coli xylose isomerase gene and its ribosome binding site was ligated into a plasmid downstream from the strong λ PL promoter. The plasmid was then used to transformE. coli strains containing a temperature-sensitive λ repressor (cI857).

Immunoaffinity purification of glucose/xylose isomerase fromStreptomyces

Applied Biochemistry and Biotechnology, 1991
A procedure was developed to purify glucose/xylose isomerase from cell extract of Streptomyces sp. NCIM 2730 using immunoaffinity chromatography. High-titer polyclonal antibodies were raised in rabbit using electrophoretically homogeneous glucose/xylose isomerase as an antigen.

Application of a compatible xylose isomerase in simultaneous bioconversion of glucose and xylose to ethanol

Biotechnology and Bioprocess Engineering, 2000
Simultaneous isomerisation and fermentation (SIF) of xylose and simultaneous isomerisation and cofermentation (SICF) of glucose-xylose mixture was carried out by the yeastSaccharomyces cerevisiae in the presence of a compatible xylose isomerase. The enzyme converted xylose to xylulose andS.

Metabolism of d-xylose in Schizosaccharomyces pombe cloned with a xylose isomerase gene

Applied Microbiology and Biotechnology, 1989
The Escherichia coli xylose isomerase gene was transformed into Schizosaccharomyces pombe for direct d-xylose utilization. In order to understand d-xylose metabolism and determine the limiting factors on d-xylose utilization by the transformed yeast, d-xylose transport, xylose isomerization, and xylulose phosphorylation were investigated.

Catabolite repression of xylose isomerase synthesis in Arthrobacter ureafaciens

Microbiology, 2008
The effect of a specific substrate as well as other carbon sources on the biosynthesis of xylose isomerase in the actinobacterium Arthrobacter ureafaciens BIM B-6 has been studied. It was established that xylose and its structural analogue xylite induced the production of the enzyme by bacterial cells.
