Combination Immunotherapy as a Promising Strategy to Overcome Immunotherapy Resistance: From Emergence to Next‐Generation Approaches

open access: yesAdvanced Science, EarlyView.
This review examines emerging combination immunotherapy strategies tailored to distinct tumor microenvironments and highlights next‐generation biomarkers that guide response prediction and treatment personalization. It integrates lessons from unsuccessful trials, addresses toxicity challenges, and outlines approaches for early biomarker discovery and ...

Tetrastigma Hemsleyanum Polysaccharide Suppresses Triple‐Negative Breast Cancer by Disrupting the Hippo‐YAP/TEAD4‐PDIA4 Axis and Endoplasmic Reticulum Stress Adaptation

open access: yesAdvanced Science, EarlyView.
ABSTRACT Triple‐negative breast cancer (TNBC) exhibits addiction to chronic endoplasmic reticulum (ER) stress, which sustains an aggressive metastatic phenotype through activation of the unfolded protein response (UPR). Here, we identify a previously unrecognized “ER‐stress addiction” axis in which the Hippo pathway effector TEAD4 directly ...

CXCR7 promoted proliferation, migration and invasion in HCC Cells by inactivating Hippo-YAP signaling

open access: yesDiscover Oncology
Background CXCR7 (ACKR3) has been well-supported as a promoter of growth and metastasis in hepatocellular carcinoma (HCC). Both CXCR7 and Hippo signaling play roles in organ development.

ER stress-induced YAP upregulation leads to chondrocyte phenotype loss in age-related osteoarthritis

open access: yesFrontiers in Pharmacology
BackgroundOsteoarthritis (OA) is a common degenerative joint disease, leading to pain and restricted mobility. Age-related endoplasmic reticulum (ER) stress has been implicated in the pathogenesis of OA, but the underlying mechanisms remain unclear. This

Activation of ERBB4 Pathway Inhibits Pathological Transdifferentiation of Lung Epithelial Progenitors into CD66c+ Basal Cells in Severe Lung Injury

open access: yesAdvanced Science, EarlyView.
In fibrotic distal lung regions, CD66c+ basal cells emerge as a pathological state. Using human distal lung organoids, this study identifies CD66c+ basal cells as a pro‐fibrotic state arising through transdifferentiation from secretory, AT2, and basal cells.

SMAD4 Palmitoylation Drives a Metabolic‐Transcriptional Circuit to Promote Tumorigenesis and Confers Radiosensitivity in Pancreatic Cancer

open access: yesAdvanced Science, EarlyView.
This study identifies palmitoylation as a novel regulatory modification of SMAD4, mediated by ZDHHC22/APT2. It activates fatty acid synthesis, creating a self‐reinforcing SMAD4–FASN–palmitate feedback loop that drives pancreatic cancer growth and enhances radiotherapy sensitivity.

Cucurbitacin E inhibits the Yes‑associated protein signaling pathway and suppresses brain metastasis of human non‑small cell lung cancer in a murine model. [PDF]

open access: yes, 2019
Human non‑small cell lung cancer (NSCLC) is associated with an extremely poor prognosis especially for the 40% of patients who develop brain metastasis, and few treatment strategies exist.

IQGAP1 participates in endothelial cell apoptosis and regulates atherosclerosis by targeting YAP.

open access: yesPLoS ONE
Endothelial cell (EC) apoptosis plays a crucial role in the onset and progression of atherosclerosis (AS). IQGAP1 is highly expressed in various tissues and affects cell growth, development and death. However, the complete elucidation of the influence of

Targeting m6A Reader YTHDF1 Enhances Antitumor Immunity and Potentiates Anti‐PD‐L1 Efficacy in Intrahepatic Cholangiocarcinoma

open access: yesAdvanced Science, EarlyView.
The m6A reader YTHDF1 drives intrahepatic cholangiocarcinoma (ICC) progression by remodeling the tumor immune microenvironment. YTHDF1 promotes MDSC recruitment via activation of m6A‐FOSL2‐CXCL6/CXCR2 axis, thereby suppressing CD8+ T cell infiltration and function.
