Protein palmitoylation: an emerging regulator of inflammatory signaling and diseases

open access: yesFrontiers in Immunology
Protein palmitoylation is a reversible lipid modification in which palmitoyl esters are covalently attached to cysteine residues of proteins. It controls various cellular physiological processes and alters protein stability, conformation, localization ...

Blueberries Reduce Palm Oil‐Induced Metabolic Endotoxemia in an In Vitro Human Intestinal‐Immune Cell Model

open access: yesMolecular Nutrition &Food Research, Volume 69, Issue 24, December 2025.
Digested palm oil and LPS treatment on intestinal Caco‐2 cells results in palm oil‐induced LPS translocation across the intestinal cell monolayer and nuclear factor kappa B (NF‐κB) activation of basolateral macrophages. Interestingly, digested whole berries reduce the palm oil + LPS‐mediated NF‐κB activation in macrophages.

Dynamic but discordant alterations in zDHHC5 expression and palmitoylation of its substrates in cardiac pathologies [PDF]

open access: yes, 2022
S-palmitoylation is an essential lipid modification catalysed by zDHHC-palmitoyl acyltransferases that regulates the localisation and activity of substrates in every class of protein and tissue investigated to date.

Breaking barriers: The cGAS‐STING pathway as a novel frontier in cancer immunotherapy

open access: yesCancer Communications, Volume 45, Issue 11, Page 1513-1546, November 2025.
Abstract Since its discovery, the cyclic GMP‐AMP synthase (cGAS)‐stimulator of the interferon gene (STING) signaling pathway has been considered a pivotal component of innate immunity and a promising target for cancer immunotherapy. Beyond its canonical role in pathogen defense, accumulating evidence has demonstrated that the cGAS‐STING pathway ...

S‑Palmitoylation during Retinoic Acid-Induced Neuronal Differentiation of SH-SY5Y Neuroblastoma Cells [PDF]

open access: yes, 2023
S-Palmitoylation is the covalent attachment of C14:0-C22:0 fatty acids (mainly C16:0 palmitate) to cysteines via thioester bonds. This lipid modification is highly abundant in neurons, where it plays a role in neuronal development and is implicated in ...

Identification of ZDHHC enzymes catalyzing acylation of influenza virus Hemagglutinin

open access: yes, 2020
Influenza viruses are negative sense segmented enveloped RNA viruses covered with 2 surface peplomers: the trimeric hemagglutinin (HA), which catalyzes virus entry by binding to sialic acid containing receptors and by performing fusion of viral with endosomal membranes and the neuraminidase (NA), which is required for release of virus particles from ...

ZDHHC12 Palmitoylates HDAC8 to Promote the Progression of Hepatocellular Carcinoma Associated with a Diet High in Saturated Fatty Acids

open access: yesAdvanced Science, Volume 12, Issue 40, October 27, 2025.
This study identifies ZDHHC12 as a critical mediator of PA‐induced HCC progression. Mechanistically, PA upregulates ZDHHC12 expression through SMARCA4 activation, leading to HDAC8 palmitoylation at Cys244 which blocks HSC70‐mediated lysosomal degradation of HDAC8, thereby promoting HCC development. These findings provide novel mechanistic insights into

ZDHHC5 Mediates Immune Dysregulation Driving Generalized Anxiety Disorder Risk

open access: yesBrain and Behavior, Volume 15, Issue 9, September 2025.
Mendelian randomization identifies palmitoylation genes driving generalized anxiety disorder (GAD) risk, notably ZDHHC5 and ZDHHC13. ZDHHC5 promotes GAD pathogenesis by modulating SSC‐A on CD4+ T cells, revealing novel immune‐mediated mechanisms and therapeutic targets for GAD.

Ubiquitination in cancer: mechanisms and therapeutic opportunities

open access: yesCancer Communications, Volume 45, Issue 9, Page 1128-1161, September 2025.
Abstract Ubiquitination, a key post‐translational modification, plays an essential role in tumor biology by regulating fundamental cellular processes, such as metabolism and cell death. Additionally, it interacts with other post‐translational modifications, which are closely linked to tumorigenesis, tumor progression, the tumor microenvironment, and ...

Zebrafish ZDHHC11 palmitoylates STING to potentiate antiviral immune response

open access: yesCell Communication and Signaling
In mammals, the activation of stimulator of interferon gene (STING), a pivotal adaptor protein mediating type I interferon (IFN-I) induction upon DNA virus infection, is regulated by palmitoylation.
