Computational Insights into β-Carboline Inhibition of Monoamine Oxidase A

open access: yesMolecules, 2022
Monoamine oxidases (MAOs) are an important group of enzymes involved in the degradation of neurotransmitters and their imbalanced mode of action may lead to the development of various neuropsychiatric or neurodegenerative disorders.

In vitro Effect of Harmine Alkaloid and Its N-Methyl Derivatives Against Toxoplasma gondii

open access: yesFrontiers in Microbiology, 2021
Toxoplasmosis is one of the most prevalent and neglected zoonotic global diseases caused by Toxoplasma gondii. The current pharmacological treatments show clinical limitations, and therefore, the search for new drugs is an urgent need in order to ...

Beyond Simple Mimicry: Next‐Generation Geometric Architectures and Future Paradigms in Small‐Molecule and Macrocyclic Peptidomimetics

open access: yesAngewandte Chemie International Edition, EarlyView.
Peptide‐to‐Small Molecule Paradigm: Peptidomimetics have evolved from simple mimicry toward drug‐like scaffolds supported by increasing clinical successes. This Perspective highlights the geometric design principles—linear repetition, convergent fusion, and cyclization—defining the next‐generation peptidomimetic architectures capable of targeting ...

Generation of the Camptothecin Scaffold by a Flavin‐Catalyzed Photooxidative Skeletal Reorganization

open access: yesAngewandte Chemie International Edition, EarlyView.
The biosynthetic pathway of the anticancer drug precursor camptothecin involves an enigmatic skeletal reorganization from a 6/5/6 tetrahydro‐β‐carboline to a 6/6/5 pyrroloquinoline ring system. Here, we show that this transformation can be achieved by photooxidation with flavin cofactors, such as flavin mononucleotide (FMN) as catalysts.

Cigarette Smoke Extract, but Not Electronic Cigarette Aerosol Extract, Inhibits Monoamine Oxidase in vitro and Produces Greater Acute Aversive/Anhedonic Effects Than Nicotine Alone on Intracranial Self-Stimulation in Rats

open access: yesFrontiers in Neuroscience, 2022
Conventional tobacco cigarettes appear to have greater abuse liability than non-combusted products such as electronic cigarettes (ECs) and nicotine replacement therapy (NRT).

The One‐Step Synthesis of Biologically Active Isochromans from Lignin‐Derived Aromatic Monomers in Tunable Deep Eutectic Solvents

open access: yesChemistry – A European Journal, EarlyView.
A straightforward one‐step protocol enables the synthesis of bio‐based and biologically active isochromans through the Oxa‐Pictet cyclization involving diverse aldehydes, including vanillin in combination with lignin‐derivable homovanillyl alcohol, a prominent aromatic platform chemical obtainable via diol‐assisted fractionation of lignocellulose ...

Dual Acid Responses of Stable Luminescent Diphenylpyridylmethyl Radicals With Additional Pyridyl Groups

open access: yesChemistry – A European Journal, EarlyView.
To expand the acid‐responsive behavior of diphenylpyridylmethyl stable luminescent radicals, additional pyridyl groups were introduced. Dual acid responsiveness was observed, involving both the newly incorporated pyridyl groups and the PyBTM moieties.

Cyclopropylamine‐Derived Distonic Iminium Radicals: Photoinduced Strategies in Chemo‐ and Stereo‐Selective Synthesis

open access: yesChemistry – A European Journal, EarlyView.
Cyclopropylamines have emerged as powerful photocatalytic building blocks. Upon light‐induced single‐electron oxidation, they undergo strain‐release ring opening to generate distonic iminium radicals that enable diverse transformations, including ring‐opening functionalization, formal [3 + 2] cycloadditions, radical cascades, and asymmetric bond ...

Acylation of indolyl subsituted β-ketonitriles. The way to new indolo[2,3-c]pyrilium and β-carboline derivatives

open access: yesЖурнал органічної та фармацевтичної хімії, 2020
Aim. To synthesize 4-cyanoindolo[2,3-c]pyrilium salts and examine their reactivity towards N-nucleophiles. Results and discussion. The acylation reaction of 2-(indolyl-3)-2-aroylacetonitriles under acid catalysis conditions was studied. 4-Cyanoindolo[2,3-
