Molecular Targets of β-Lactam-Based Antimicrobials : Beyond the Usual Suspects
The common practice in antibacterial drug development has been to rapidly make an attempt to find ever-more stable and broad-spectrum variants for a particular antibiotic, once a drug resistance for that antibiotic is detected.

A biohybrid nanorobot integrating lytic bacteriophage N4 with Pd nanozymes is developed for targeted eradication of multidrug‐resistant E. coli biofilms. Synergistic bacterial lysis and ROS‐mediated oxidation enable simultaneous biofilm removal and antibiotic resistance genes degradation, maintaining high efficacy in complex wastewater environments ...

By leveraging this homodimerization mechanism, molecular glues were rationally designed to induce dysfunctional 3A dimerization, thereby restoring antiviral RNAi. The optimal molecular glue, VTP‐32, demonstrated potent and pan‐enterovirus (groups A, B, D) antiviral effects.

A pathogen‐centric antibody‐antibiotic conjugate (AZO‐AAC) releases antibiotics via bacterial azoreductase, independent of host‐lysosomal function. This mechanism achieves nanomolar eradication of both planktonic and intracellular methicillin‐resistant Staphylococcus aureus (MRSA).

Deletion of Lytic Transglycosylases Increases Beta-Lactam Resistance in Shewanella oneidensis
Production of chromosome-encoded β-lactamases confers resistance to β-lactams in many Gram-negative bacteria. Some inducible β-lactamases, especially the class C β-lactamase AmpC in Enterobacteriaceae, share a common regulatory mechanism, the ampR-ampC ...

Phylogenetic and biochemical analyses of the heme transporter CydDC reveal its functional conservation throughout bacterial evolution and demonstrate its unique asymmetric allosteric mechanism. Furthermore, impairment of CydDC function directly affects bacterial antibiotic resistance and likely compromises antibiotic efficacy through drug efflux.

Comparative Evaluation of the in-vitro Activity of Six β-lactam/β-lactamase Inhibitor Combinations against Gram Negative Bacilli [PDF]
Background: The extensive use of the β-lactam antibiotics in hospitals and in the community has created major resistance problems which has led to increased morbidity, mortality and healthcare costs.

Trimeric Tobramycin/Nebramine Synergizes β‑Lactam Antibiotics against Pseudomonas aeruginosa
β-Lactam antibiotics remain one of the most effective therapeutics to treat infections caused by Gram-negative bacteria (GNB). However, since ancient times, bacteria have developed multiple resistance mechanisms toward this class of antibiotics including
