Drug Discovery in the Field of β-Lactams: An Academic Perspective

open access: yesAntibiotics
β-Lactams are the most widely prescribed class of antibiotics that inhibit penicillin-binding proteins (PBPs), particularly transpeptidases that function in peptidoglycan synthesis.

Antibiotic Remodeling of the Gut Microbiota Impairs Host Resistance to Secondary Infection

open access: yesiMetaMed, EarlyView.
Gut microbial dynamics induced by four antibiotics, revealed distinct taxonomic and functional shifts. Vancomycin‐perturbed bacteriomes were proven to promote pathogen resistance to antibiotic treatment. Metronidazole‐treated mice showed pronounced Salmonella translocation to hepatic tissue, and altered Enterococcus avium populations modulated host ...

Comparative Evaluation of the in-vitro Activity of Six β-lactam/β-lactamase Inhibitor Combinations against Gram Negative Bacilli [PDF]

open access: yesJournal of Clinical and Diagnostic Research, 2013
Background: The extensive use of the β-lactam antibiotics in hospitals and in the community has created major resistance problems which has led to increased morbidity, mortality and healthcare costs.

Molecular Basis of Bicyclic Boronate β-Lactamase Inhibitors of Ultrabroad Efficacy – Insights From Molecular Dynamics Simulation Studies

open access: yesFrontiers in Microbiology, 2021
β-Lactam antibiotics represent about 70% of all antibacterial agents in clinical use. They are safe and highly effective drugs that have been used for more than 50 years, and, in general, well tolerated by most patients.

Evaluation of Ceftriaxone as a Monotherapy and Amoxicillin With Clavulanic Acid as Combination Therapy in Surgical Prophylaxis

open access: yesiNew Medicine, EarlyView.
ABSTRACT Surgical antibiotic prophylaxis (SAP) is crucial in preventing postoperative surgical site infections (SSIs), especially in clean and clean‐contaminated surgeries. It was a prospective comparative observational study that compared the effectiveness and safety of single‐dose ceftriaxone monotherapy with amoxicillin–clavulanic acid as a ...

Potent monoclonal antibodies against multidrug‐resistant hypervirulent Klebsiella pneumoniae

open access: yesInterdisciplinary Medicine, EarlyView.
A novel immunization strategy using a low‐virulence, multidrug‐resistant strain yields synergistic monoclonal antibodies against hypervirulent Klebsiella pneumoniae. These antibodies provide cross‐serotype protection through a dual‐mechanism of pathogen clearance and immunomodulation, offering a promising non‐antibiotic therapeutic for resistant ...

Use of β-Lactam/β-Lactamase Inhibitors for Extended-Spectrum-β-Lactamase Infections: Defining the Right Patient Population

open access: yes, 2017
In a multicenter, multinational observational study that included neutropenic patients with bloodstream infections by extended-spectrum-β-lactamase-producing species, Gudiol et al. (Antimicrob. Agents Chemother.

Duplex Recombinase Polymerase Amplification‐Lateral Flow Assay for Rapid Visual Detection of Critical Antibiotic Resistance Genes in Escherichia coli and Klebsiella pneumoniae From Positive Blood Cultures

open access: yesJournal of Clinical Laboratory Analysis, EarlyView.
This study developed a pair of duplex RPA‐LFA assays for the simultaneous detection of critical resistance genes, blaCTX‐M/mcr‐1 and blaoxa‐48‐like/blaNDM, from hemoculture samples. Under optimized conditions, duplex RPA was performed at 37°C–38°C for 25 min, followed by LFA for 10–15 min.

Bicyclic Boronates as Potent Inhibitors of AmpC, the Class C β-Lactamase from Escherichia coli

open access: yesBiomolecules, 2020
Resistance to β-lactam antibacterials, importantly via production of β-lactamases, threatens their widespread use. Bicyclic boronates show promise as clinically useful, dual-action inhibitors of both serine- (SBL) and metallo- (MBL) β-lactamases.

On‐site pilot‐scale advanced oxidation pretreatment of hospital wastewater: assessment of pharmaceuticals, pathogens, antibiotic resistance genes and acute ecotoxicity

open access: yesJournal of Chemical Technology &Biotechnology, EarlyView.
Abstract BACKGROUND Hospital premises generate wastewater containing a wide spectrum of contaminants, such as pharmaceutically active compounds (PhACs), antibiotic resistance genes (ARGs) and pathogenic microorganisms, whilst plenty of them are classified under the antibiotic resistant bacteria (ΑRΒ) category.
