HPLC characterization, molecular docking, QSAR and molecular dynamics simulation of Alstonia boonei phytochemical analogs as potent 5-alpha reductase inhibitors

open access: yesDiscover Chemistry
Background Current chemotherapy for Benign Prostatic Hyperplasia (BPH) relies on 5-alpha reductase inhibitors like Finasteride and Dutasteride, which are associated with significant side effects.

Nitrogen‐Boosted H2O2 Production of Arginine‐Polyphenol Nanozyme Drives Oxidative Eustress for Hair Regeneration

open access: yesAdvanced Science, EarlyView.
PEA nanoenzymes are assembled from L‐arginine and EGCG. The nitrogen element in arginine enhances the thermodynamic feasibility of H2O2 production through PEA's oxygen reduction catalysis, enabling nitrogen‐dependent H2O2 release. Intradermal delivery of PEA via microneedles promotes efficient H2O2 synthesis, thereby stimulating regrowth in telogen ...

Hormonal manipulation of lower urinary tract symptoms secondary to benign prostatic obstruction

open access: yesIndian Journal of Urology, 2014
Although the etiology of lower urinary tract symptoms (LUTS) is often multifactorial, a significant proportion of men over the age of 50 suffer from benign prostatic obstruction (BPO) secondary to benign prostatic hyperplasia. Prostate, being an androgen

Selenoprotein H Functions as a PPARα Coactivator to Link Selenium Homeostasis to Hepatic Lipid Metabolism and Protect against Steatohepatitis

open access: yesAdvanced Science, EarlyView.
Our study identifies selenium deficiency as a hallmark of MASH pathogenesis. Dietary selenium supplementation enhances hepatic fatty acid oxidation (FAO) and attenuates MASH progression by activating the PPARα pathway via selenoprotein H (SELENOH). This selenium‐SELENOH‐PPARα nexus redefines the functional scope of selenoproteins, moving from redox ...

Comparative Effectiveness and Safety of Finasteride and Dutasteride in the Treatment of Benign Prostatic Hyperplasia: A Real-World Retrospective Study

open access: yesMedicina
Background and Objectives: Benign prostatic hyperplasia (BPH) is one of the most common chronic conditions in older men, significantly impairing quality of life (QoL) by causing lower urinary tract symptoms (LUTSs). 5-alpha-reductase inhibitors (5-ARIs),

Engineering New‐to‐Nature Biological Pathways for β,γ‐Alkanediol Synthesis

open access: yesAdvanced Science, EarlyView.
β,γ‐Alkanediols are value‐added chemicals to be used as functional solvents, biofuels, and cosmetic components. This work demonstrates a carboligation‐mediated approach for synthesis of linear chain β,γ‐alkanediols in Escherichia coli, namely, hexane‐2,3‐diol (2,3‐HDO) and pentane‐2,3‐diol (2,3‐PDO). The engineered E. coli cells produce 152.2 mm (17.98

Neuronal Cholesterol Deficiency Mediated by Astrocytic SREBP2 Downregulation Leads to Postoperative Cognitive Dysfunction Through Impairment of Hippocampal Synaptic Plasticity and Excitatory Synaptic Transmission

open access: yesAdvanced Science, EarlyView.
The results showed that the reduction of plasma 24‐hydroxycholesterol (24‐OHC) was related to the cognitive decline in elderly surgical patients. Neuroinflammation induced by anesthesia and surgery significantly downregulated SREBP2 in hippocampal reactive astrocytes, resulting in reduced cholesterol synthesis.

Bilirubin as a Modulator of WNK1 Protein Signaling: Implications for Neuroinflammatory Diseases

open access: yesAdvanced Science, EarlyView.
ABSTRACT Previously regarded merely as a potentially harmful waste product of heme catabolism, bilirubin has now emerged as a pleiotropic molecule with potent antioxidant, anti‐inflammatory, and hormone‐like properties. Recent findings have revealed protective effects against cardiovascular, metabolic, autoimmune, and neoplastic diseases, as well as ...

The efficacy of topical minoxidil 2% versus topical botanically derived inhibitors of 5 alpha reductase in treatment of female pattern hair loss by trichoscopy

open access: yesJournal of the Egyptian Women’s Dermatologic Society, 2019
Background Female pattern hair loss (FPHL) is a distressing common problem. Minoxidil is the only FDA approved medical treatment for FPHL. However, the off-label botanically derived 5-alpha reductase inhibitors (5αRIs) are commonly used despite ...

T Cell Exhaustion in Cancer Immunotherapy: Heterogeneity, Mechanisms, and Therapeutic Opportunities

open access: yesAdvanced Science, EarlyView.
T cell exhaustion limits immunotherapy efficacy. This article delineates its progression from stem‐like to terminally exhausted states, governed by persistent antigen, transcription factors, epigenetics, and metabolism. It maps the exhaustion landscape in the TME and proposes integrated reversal strategies, providing a translational roadmap to overcome
