Role of ALKBH1 in the Core Transcriptional Network of Embryonic Stem Cells

open access: yesCellular Physiology and Biochemistry, 2016
Background/Aims: ALKBH1, an AlkB homologue in the 2-oxoglutarate and Fe2+ dependent hydroxylase family, is a histone dioxygenase that removes methyl groups from histone H2A. Studies of transgenic mice lacking Alkbh1 reveal that most Alkbh1-/- embryos die

Arsenic regulates ALKBH1 abundance and substrate specificity to promote translation and tumorigenicity

open access: yesCell Reports
Summary: The mechanisms that underlie arsenic tumorigenicity remain incompletely understood. Here, we show that arsenic regulates the abundance and the demethylase activity of ALKBH1, which, in turn, promotes arsenic-induced skin tumorigenesis.

ALKBH1 gene polymorphisms confer hepatoblastoma susceptibility in Chinese children

open access: yesScientific Reports
Hepatoblastoma is the most common malignant liver tumor of embryonic origin in children; however, its underlying etiology remains unclear. N1-methyladenosine (m1A) modification is widely found in various human tissues and has been demonstrated to ...

ALKBH1 activity in vitro and human cell lines by isotope dilution mass spectrometry.

open access: yesPLoS ONE
AlkB homolog 1 (ALKBH1) is a member of the AlkB family of Fe(II) and α-ketoglutarate (α-KG)-dependent dioxygenases, known for its enzymatic activity on various nucleic acid substrates.

Spatial and single-cell analyses uncover links between ALKBH1 and tumor-associated macrophages in gastric cancer

open access: yesCancer Cell International
Background AlkB homolog 1, histone H2A dioxygenase (ALKBH1), a crucial enzyme involved in RNA demethylation in humans, plays a significant role in various cellular processes.

YTHDF3 recognizes DNA N6-methyladenine and recruits ALKBH1 for 6mA removal from genomic DNA

open access: yesThe EMBO Journal
DNA N6-methyladenine (6mA) is an emerging epigenetic mark in the mammalian genome. ALKBH1 preferentially exhibits 6mA demethylase activity for single-stranded DNA (ssDNA) or bubbled/bulged DNA, but not for double-stranded DNA (dsDNA).

ALKBH1 Gene rs6494 T>A Polymorphism Decreases Wilms Tumour Risk in Chinese Children

open access: yesJournal of Cellular and Molecular Medicine, Volume 29, Issue 18, September 2025.
ABSTRACT Wilms tumour (WT) is the most predominant renal carcinoma that affects children, and the understanding of the genetic mechanisms underlying WT development is continually evolving. The role of the demethylase ALKBH1, which is known for its association with diverse cancers, in WT has never been explored.

ALKBH1 is an RNA dioxygenase responsible for cytoplasmic and mitochondrial tRNA modifications [PDF]

open access: yesNucleic Acids Research, 2017
ALKBH1 is a 2-oxoglutarate- and Fe2+-dependent dioxygenase responsible for multiple cellular functions. Here, we show that ALKBH1 is involved in biogenesis of 5-hydroxymethyl-2΄-O-methylcytidine (hm5Cm) and 5-formyl-2΄-O-methylcytidine (f5Cm) at the first position (position 34) of anticodon in cytoplasmic tRNALeu, as well as f5C at the same position in
