Targeting the MDM2‐MDM4 interaction interface reveals an otherwise therapeutically active wild‐type p53 in colorectal cancer

open access: yesMolecular Oncology, EarlyView.
This study investigates an alternative approach to reactivating the oncosuppressor p53 in cancer. A short peptide targeting the association of the two p53 inhibitors, MDM2 and MDM4, induces an otherwise therapeutically active p53 with unique features that promote cell death and potentially reduce toxicity towards proliferating nontumor cells.

Primary extranodal diffuse large B‐cell lymphoma: Molecular features, treatment, and prognosis

open access: yesAging and Cancer, Volume 3, Issue 3-4, Page 133-146, September-December 2022., 2022
One‐third of DLBCL arises from extranodal organs and is challenging to manage. Molecular features are critical to elucidate the differences in clinical features, predict the disease prognosis, and improve effective targeted therapeutic strategies. Abstract Diffuse large B‐cell lymphoma (DLBCL) is the most common subtype of non‐Hodgkin's lymphoma and ...

Thermal proteome profiling and proteome analysis using high‐definition mass spectrometry demonstrate modulation of cholesterol biosynthesis by next‐generation galeterone analog VNPP433‐3β in castration‐resistant prostate cancer

open access: yesMolecular Oncology, EarlyView.
Elevated level of cholesterol is positively correlated to prostate cancer development and disease severity. Cholesterol‐lowering drugs, such as statins, are demonstrated to inhibit prostate cancer. VNPP433‐3β interrupts multiple signaling and metabolic pathways, including cholesterol biosynthesis, AR‐mediated transcription of several oncogenes, mRNA 5′

Holistic structure of neural pathways underlies brain perceptual rivalry: Physical mechanism of auditory stream segregation [PDF]

open access: yesarXiv
Brain perceptual rivalry, exemplified by auditory stream segregation of competing tones (A_, B__, ABA_), serves as a core mechanism of brain perception formation. While increasingly recognized as determining by neural connections rather than specific neural groups, the mechanism of brain perception remains uncertain. We demonstrate that auditory stream

Iterative variational learning of committor-consistent transition pathways using artificial neural networks [PDF]

open access: yesarXiv
This contribution introduces a neural-network-based approach to discover meaningful transition pathways underlying complex biomolecular transformations in coherence with the committor function. The proposed path-committor-consistent artificial neural network (PCCANN) iteratively refines the transition pathway by aligning it to the gradient of the ...
