L‐aspartate oxidase provides new insights into fumarate reduction in anaerobic darkness in Synechocystis sp. PCC6803

open access: yesFEBS Letters, EarlyView.
Synechocystis strains deficient in succinate dehydrogenase (SDH) secrete more succinate than the WT under dark anaerobic conditions, supporting that SDH then primarily acts as SDH, not as a fumarate reductase. L‐aspartate oxidase (Laspo) from Synechocystis is functional under anaerobic conditions, reducing fumarate to succinate.

In vitro 3D skin culture and its sustainability in toxicology: a narrative review

open access: yesArtificial Cells, Nanomedicine, and Biotechnology
In current toxicological research, 2D cell cultures and animal models are well- accepted and commonly employed methods. However, these approaches have many drawbacks and are distant from the actual environment in human.

Leucine‐rich glioma inactivated 1 (LGI1) is a ganglioside‐binding protein

open access: yesFEBS Letters, EarlyView.
Neuronal hyperexcitability associated with a decrease/absence of the extracellular protein LGI1 has been suggested to be primarily due to the downregulation of Kv1 channel expression. The molecular mechanisms underlying this decrease have not yet been elucidated.

Development of a predictive in vitro test module for developmental neurotoxicity testing using mouse embryonic stem cells

open access: yes, 2008
The development of the nervous system is a rather complex process known to be affected by different drugs and chemicals. Therefore, regulatory test guidelines have been adopted for the prediction and assessment of developmental neurotoxicity (U.S.EPA ...

Implementation of the 3Rs Concept in Quality Control of Biologicals: Status and Prospects (Review)

open access: yesБезопасность и риск фармакотерапии
INTRODUCTION. Many countries are implementing an increasing number of alternatives to animal testing in the research, development, and quality control of medicines under the concept of 3Rs (replacement, reduction, and refinement).

Metastatic niche shaped by host factors influences disseminated cancer cell fate

open access: yesFEBS Letters, EarlyView.
Metastasis is shaped not only by cancer cells but also by the environments they encounter. This review explores how factors such as aging, diet, the microbiome, lifestyle, and environmental exposures remodel organ‐specific niches in the lung, liver, bone, and brain, influencing where metastatic cells survive, remain dormant, or grow, and ultimately ...

Go3R - intelligent internet searches for alternatives to animal testing

open access: yes, 2010
EU Directive 86/609/EEC for the protection of laboratory animals obliges scientists to consider whether a planned animal experiment can be replaced, reduced or refined.

A novel human organotypic tumor invasion model of cell line MDA-MB-231 in Precision-Cut Lung Slices (PCLS) to reduce animal experiments in preclinical oncology

open access: yes, 2022
S.134Cancer such as breast carcinoma is a major public health problem worldwide. The actual cause of death is the formation of metastasis which often occurs in the lung.

Membrane composition and thermodynamic identity as boundaries of life for synthetic cell research

open access: yesFEBS Letters, EarlyView.
What makes a cell a cell? The boundary of a living cell is not just a wall. Read as a Markov blanket, the membrane separates internal from external states, generating identity and non‐equilibrium order. Can this identity be rebuilt from scratch in a synthetic cell?

KDM7A and KDM1A inhibition suppresses tumour promoting pathways in prostate cancer

open access: yesMolecular Oncology, EarlyView.
Treatment resistance is a major challenge for patients with advanced prostate cancer. This study examined an alternative approach to target the major prostate cancer‐promoting pathway by targeting epigenetic factors, whose levels are higher in tumours.
