Conformation and toxicity of amanitins

Amanitin (AMA) is responsible for human fatalities after ingestion of poisonous mushrooms, thus, a rapid and accurate detection method is urgently needed. Here, gold nanoparticle-based immunosensor with monoclonal antibody against AMA was constructed for rapid detection of alpha- and beta-amanitin (α- and β-AMA) in mushroom, serum and urine samples ...

Archives of Virology, 1978
The advances in virus research made possible by the use of amanitins are reported; they include i) determination of the genetic source (i.e. cellular or viral) of the RNA polymerase transcribing the viral genome, and ii) mechanism of virus-induced shut-off of host RNA synthesis.

Viroid replication is inhibited by α-amanitin
Nature, 1979VIROIDS are infectious single-stranded circular RNA molecules1 which cause disease in higher Plants. Although the complete molecular structure of one viroid has recently been established2, the mechanism of viroid replication and patho-genesis is still unknown.

Abstract 735: SAR of amanitin and optimization of linker-amanitin derivatives for solid tumors
Cancer Research, 2018Abstract Background: ATACs (Antibody Targeted Amanitin Conjugates) comprise a new class of antibody-drug conjugates using amanitin as toxic payload. Amanitin binds to the eukaryotic RNA pol II and thereby inhibits the cellular transcription process at very low concentrations.

Amanitin binding to RNA polymerase II in α-amanitin-resistant rat myoblast mutants
Journal of Molecular Biology, 1977A series of independent α-amanitin-resistant (Ama R ) mutants have been isolated from diploid and tetraploid strains of L6 rat myoblast cells. With one exception, these mutants contain both α-amanitin-sensitive wild-type and α-amanitin-resistant mutant RNA polymerase II activities.

α-Amanitin: Inactivation by bovine lactoperoxidase
Experientia, 1979The principle amatoxin, alpha-amanitin, is found to be extremely sensitive toward lactoperoxidase catalyzed degradation, rather than iodination, of the indole nucleus. Extensive attenuation of inhibitor potency against eukaryotic DNA-dependent RNA polymerase II accompanies the treatment of alpha-amanitin with lactoperoxidase, iodide and hydrogen ...

Total Synthesis of α‐ and β‐Amanitin
Angewandte Chemie International Edition, 2020Abstractα‐Amanitin and related amatoxins have been studied for more than six decades mostly by isolation from death cap mushrooms. The total synthesis, however, remained challenging due to unique structural features. α‐Amanitin is a potent inhibitor of RNA polymerase II. Interrupting the basic transcription processes of eukaryotes leads to apoptosis of

Antidotes to Experimental cc-Amanitin Poisoning
Nature New Biology, 1972THE two most important toxic cyclopeptides isolated from the mushroom Amanita phalloides (death cap) are phalloidin and α-amanitin1, 2. The latter is considered to be responsible for the fatal cases of poisoning. Up to now, protection has been demonstrated either with a non-toxic chemical analogue (antamanide) isolated from the toadstool itself and ...
