Life in vacuoles – nutrient acquisition by Leishmania amastigotes

International Journal for Parasitology, 2001
Leishmania have a digenetic life cycle, involving a motile, extracellular stage (promastigote) which parasitises the alimentary tract of a sandfly vector. Bloodfeeding activity by an infected sandfly can result in transmission of infective (metacyclic) promastigotes to mammalian hosts, including humans. Leishmania promastigotes are rapidly phagocytosed

Heme as a source of iron to Leishmania infantum amastigotes

Acta Tropica, 2009
Amastigotes, the mammalian stage of Leishmania, must acquire iron from molecules accessing the macrophage parasitophorous vacuole (PV) where they inhabit. These molecules likely include non-heme and heme-bound forms of iron. Here we demonstrate that, in addition to the previously documented use of ferrous iron, Leishmania amastigotes are also capable ...

Leishmania mexicana: Energy metabolism of amastigotes and promastigotes

Experimental Parasitology, 1982
Abstract The utilisation of substrates by Leishmania mexicana amastigotes and promastigotes differed significantly. The rates of uptake and catabolism of nonesterified fatty acids were up to 10-fold higher with amastigotes. Almost all the available exogenous fatty acids were consumed during amastigote transformation and by stationary phase of ...

Concanavalin a Binding Receptors on Trypanosoma cruzi Amastigotes

The Journal of Parasitology, 1980
501-505) as adapted by Garcia et al. (1978, Arch. Biochem. Biophys. 188: 315-322). The bugs used in our experiments consumed 160 to 200 mg of blood. Only 15% of the insects did not feed fully and were discarded. The number of bugs dissected at different intervals after infection and the number of parasites per bug are summarized in Table I.

Migration of Leishmania donovani Amastigotes in the Cerebrospinal Fluid

The American Journal of Tropical Medicine and Hygiene, 1996
A 10-year-old boy had been suffering from kala-azar (visceral leishmaniasis) for two and a half years. He failed to respond to all known anti-leishmanial treatment regimens. Even the drastic step of splenectomy failed to cure him. In November 1991, he presented with symptoms of meningitis.

Leishmania enriettii Amastigotes: Demonstration of Fibrillar Surface

The Journal of Infectious Diseases, 1982
An extensive fibrillar surface was identified in situ by transmission electron microscopy on the amastigote stage of the macrophage intracellular parasite, Leishmania enriettii. Attachment to the amastigote surface of 20-160-nm fibrils was clearly evident on organisms that had been isolated from guinea pigs.

Effect of Gossypol on Trypomastigotes and Amastigotes of Trypanosoma cruzi

The Journal of Protozoology, 1990
ABSTRACTBloodstream Trypanosoma cruzi trypomastigotes isolated from infected mice undergo reduction of motility and structural damages after 5 to 45 min exposure to gossypol at concentrations ranging from 5 to 50 μM. When 1% serum albumin is added to the incubation medium, no alterations of parasites are observed, even with 100 μM gossypol ...

An antigenically distinct lipophosphoglycan on amastigotes of Leishmania major

Molecular and Biochemical Parasitology, 1991
We show that lipophosphoglycan (LPG) on the surface of amastigotes of Leishmania major is antigenically and biochemically distinct from promastigote LPG. A rabbit antiserum raised against the amastigote integral membrane fraction detected LPG spanning the region of Mr 55,000-100,000 on Western blots of the amastigote integral membrane fraction, but did

Leishmania tropica: Differences in the antigenicity of promastigotes and amastigotes

Cellular Immunology, 1985
In vitro bioassays were used to compare T-cell responses induced by the intramacrophage amastigote stage and the sandfly-borne promastigote stage of Leishmania tropica. Lymph node cells from mice immunized with sonicated promastigotes or amastigotes incorporated in Freund's complete adjuvant were assayed for their ability to proliferate and to release ...

Long Term Cryopreservation of the Amastigote Stages of Hemoflagellates

The Journal of Protozoology, 1988
ABSTRACT. Tissue homogenates containing amastigotes of either Leishmania donovani, L. tropica, or Trypanosoma cruzi were rapidly frozen with 10% glycerol as cryoprotectant. Viability and pathogenicity were maintained for at least 23 years with the Khartoum strain of L. donovani, 22 years with the Malakal strain of L.
