Detection of lectin activity in Leishmania promastigotes and amastigotes

Cell lysates from 16 strains of eight Leishmania species were used to test haemagglutination activity (HA) against a variety of RBC. HA was detected using native or neuraminidase-treated rabbit RBC; it was found in promastigotes of all the Leishmania strains tested and in axenic amastigotes of L. mexicana.

Following intravenous (i.v.) injection of formalin-killed or ultraviolet-killed or ultraviolet-irradiated amastigotes of Leishmania donovani, the parasites could be identified in mouse liver at 5 min, but most amastigotes were digested by 3 hr or between 24 and 72 hr, respectively.

Purine metabolism in Leishmania donovani amastigotes was found to be similar to that of promastigotes with the exception of adenosine metabolism. Adenosine kinase activity in amastigotes is approximately 50-fold greater than in promastigotes. Amastigotes deaminate adenosine to inosine through adenosine deaminase, an enzyme not present in promastigotes.
