Malaria is the protozoal disease most feared by travellers to the tropics, but other protozoal diseases such as leishmaniasis, trypanosomiasis, amoebiasis and giardiasis cause havoc for many millions of people.

The therapeutic potential of inhibitors of dihydrofolate reductase as selectively toxic agents was first realized in early studies involving laboratory species of the genus Plasmodium. These experiments were carried out initially with readily standardized infections of Plasmodium gallinaceum in chicks and P. berghei in mice.
