Facile conversion of RNA aptamers to modular fluorescent sensors with tunable detection wavelengths. [PDF]

open access: yes, 2011
A GTP aptamer was converted to a modular fluorescent GTP sensor by conjugation of RRE (Rev responsive element) RNA and successive complex formation with a fluorophore-modified Rev peptide.

Programming Next‐Generation Synthetic Biosensors by Genetic Circuit Design

open access: yesAdvanced Science, EarlyView.
Synthetic biology enables genetic circuit‐based biosensing to detect diverse targets, process signals, and transduce them into readable outputs or intracellular regulatory activities. However, field deployment and real‐world application of such synthetic biosensors face considerable challenges in sensitivity, specificity, speed, stability, and ...

Ultrafast Capillary Electrophoresis Isolation of DNA Aptamer for the PCR Amplification-Based Small Analyte Sensing

open access: yesFrontiers in Chemistry, 2015
Here, we report a new homogeneous DNA amplification-based aptamer assay for small analyte sensing. The aptamer of adenosine chosen as the model analyte was split into two fragments able to assemble in the presence of target.

A Solution for Exosome‐Based Analysis: Surface‐Enhanced Raman Spectroscopy and Artificial Intelligence

open access: yesAdvanced Intelligent Discovery, EarlyView.
Exosomes are emerging as powerful biomarkers for disease diagnosis and monitoring. This review highlights the integration of surface‐enhanced Raman spectroscopy with artificial intelligence to enhance molecular fingerprinting of exosomes. Machine learning and deep learning techniques improve spectral interpretation, enabling accurate classification of ...

RNA aptamer delivery through intact human skin [PDF]

open access: yes, 2018
It is generally recognised that only relatively small molecular weight (typically 100,000-fold) and aptamer integrity was confirmed using an oligonucleotide precipitation assay.

Understanding Neurodegenerative Diseases From the −Omics Perspective: Lessons Learnt

open access: yesAnnals of Neurology, EarlyView.
As the population ages, certain neurodegenerative diseases (NDs) are becoming a major health issue. For this reason, this review will focus on the most common ND with onset after 65 years old; Alzheimer's disease, Parkinson's disease, Lewy body dementia, and frontotemporal dementia.

Mucin Glycoprotein Nanoparticles Enable a Selective Antisense Therapy for Oncogenic MicroRNAs

open access: yesAdvanced NanoBiomed Research, EarlyView.
Mucin glycoproteins are turned into nanoparticles by employing synthetic DNA strands, which have a dual function: they stabilize the nanoparticles and act as binding sites for intracellular miRNA‐21. Thus, upon internalization into tumor cells, these mucin nanoparticles can deplete miRNA‐21 from the cytosol, which induces apoptosis in vitro and in vivo.

Developments in the tools and methodologies of synthetic biology. [PDF]

open access: yes, 2014
Synthetic biology is principally concerned with the rational design and engineering of biologically based parts, devices, or systems. However, biological systems are generally complex and unpredictable, and are therefore, intrinsically difficult to ...

In vitro evolution of chemically-modified nucleic acid aptamers: Pros and cons, and comprehensive selection strategies

open access: yesRNA Biology, 2016
Nucleic acid aptamers are single-stranded DNA or RNA oligonucleotide sequences that bind to a specific target molecule with high affinity and specificity through their ability to adopt 3-dimensional structure in solution.
