Rheumatoid arthritis is a disease characterized by joint inflammation. Approximately 50% of patients show insufficient response to traditional synthetic disease‐modifying antirheumatic drugs. This study aims to elucidate differential molecular profiles of the mechanisms underlying drug responses through multi‐omics strategy.

Structural insight into the ligand binding mechanism of aryl hydrocarbon receptor
The aryl hydrocarbon receptor (AHR) plays important roles in regulating xenobiotic metabolism, cellular differentiation, and immunity. Here, the authors present multiple crystal structures of the Drosophila AHR PAS-B domain, providing insights into the ...

Decoding Human Placental Cellular and Molecular Responses to Obesity and Fetal Growth
Women with obesity often deliver large‐for‐gestational‐age (LGA) infants. Single‐nucleus RNA sequencing of term placenta reveals that hypoxia and TNF‐α signaling in syncytiotrophoblasts are featured in maternal obesity, but inflammatory signatures in Hofbauer cells and response to lipid or carbohydrate metabolism in fibroblasts are specific to LGA.

Solid predominant lung adenocarcinoma exhibits an immune‐excluded, ferroptosis‐resistant niche enriched with IL4I1⁺ TAMs and TDO2⁺ myCAFs. Spatial and multi‐omics analyses reveal AhR‐driven crosstalk that promotes T cell exhaustion and therapy resistance. Blocking AhR with CH‐223191 restores ferroptosis sensitivity, and its combination with ferroptosis

AIP limits neurotransmitter release by inhibiting calcium bursts from the ryanodine receptor
In this work the authors show that AIP (aryl hydrocarbon receptor-interacting protein), a gene frequently mutated in association with pituitary tumors, regulates neurotransmitter release by inhibiting calcium release from ryanodine receptors.

The aryl hydrocarbon receptor as a model PAS sensor
Proteins containing PER-ARNT-SIM (PAS) domains are commonly associated with environmental adaptation in a variety of organisms. The PAS domain is found in proteins throughout Archaea, Bacteria, and Eukarya and often binds small-molecules, supports ...

A solid‐state fermented probiotic (PYW) is developed using wheat bran as substrate, containing high viable Lacticaseibacillus paracasei and bioactive metabolites. PYW alleviates intestinal inflammation by gut microbiota composition, enriching indole derivatives, activating aryl hydrocarbon receptor signaling, and regulating macrophage polarization. PYW
