“Membrane‐Guided” Repair Strategy: Precision Delivery of GGT1 Degrader for Targeted Repair and Regeneration of Spinal Cord Neurons

open access: yesAdvanced Science, EarlyView.
This study confirms that GGT1 is a key driver of neuronal ferroptosis following spinal cord injury. We developed NSCm@EA, a biomimetic delivery system coated with neural stem cell membranes, for precise delivery of enocyanin to injured neurons. By combining targeted delivery with ubiquitination degradation mechanisms, this system promotes MGRN1 ...

PANX2 Suppresses Lung Adenocarcinoma Progression by Inducing Disulfidptosis and Enhancing Antitumor Immunity

open access: yesAdvanced Science, EarlyView.
ABSTRACT Lung adenocarcinoma (LUAD) remains a leading cause of cancer mortality with limited therapeutic options. Disulfidptosis, a novel cell death modality driven by disulfide stress, represents a promising target, yet its regulation in LUAD is poorly defined. Here, we identify Pannexin 2 (PANX2) as a tumor suppressor in LUAD.

PGK1 Suppresses CD8+ T Cell‐Mediated Antitumor Immunity Through CCL2/CCR2/Tumor‐Associated Macrophages Axis in Hepatocellular Carcinoma

open access: yesAdvanced Science, EarlyView.
PGK1 in tumor cells upregulates CCL2 expression through activation of the AKT/GSK‐3β/β‐catenin signaling axis, thereby promoting the recruitment and M2 polarization of TAMs and ultimately impairing the infiltration and activation of CD8+ T cells within the HCC tumor microenvironment. ABSTRACT Patients with advanced hepatocellular carcinoma (HCC) have a

mTORC2 Phosphorylation of GSDME‐N Drives Cullin4B‐Mediated Proteasomal Degradation to Suppress Pyroptosis and Confer Radioresistance in Small Cell Lung Cancer

open access: yesAdvanced Science, EarlyView.
Radioresistance severely limits the efficacy of therapies for small cell lung cancer (SCLC). This study reveals a novel mechanism of resistance driven by the active suppression of pyroptosis. Specifically, the mTORC2 complex directly phosphorylates GSDME‐N and promotes its CUL4B‐mediated ubiquitination and proteasomal degradation.

Identification and validation of endoplasmic reticulum autophagy-related potential biomarkers in periodontitis

open access: yesScientific Reports
Periodontitis is a chronic inflammatory disease that mainly occurs in the dental supporting tissues. Endoplasmic reticulum autophagy (ER-phagy) is a new type of selective autophagy. The main function of ER-phagy is to degrade excess ER membranes or toxic protein aggregates, control the volume of ER, and maintain cell homeostasis.

Synergistic Remodeling of Tumor Immune Microenvironment via a DNA Nanodevice Integrating STING Activation and Lysosome‐Targeted PD‐L1 Degradation

open access: yesAdvanced Science, EarlyView.
A rational design DNA nanoplatform not only achieves efficient PD‐L1 degradation but also triggers robust STING signaling. The nanodevice effectively reprograms “cold” tumors, leading to potent inhibition of tumor growth and metastasis in vivo. ABSTRACT The cGAS‐STING pathway is a cornerstone of innate antitumor immunity; however, its therapeutic ...

Multi-omics analysis and validation of autophagy-related diagnostic biomarker in osteoarthritis

open access: yesAnnals of Medicine
Autophagy plays an essential regulatory role in the development of osteoarthritis (OA), its specific regulatory mechanism remains unclear. This work aims to study the critical genes related to autophagy in OA and explore their potential value. Datasets were downloaded from the publicly available GEO database, and differentially expressed autophagy ...

Imaging‐Guided Live Single‐Cell Lipid Profiling of Leader and Follower Cellsduring Collective Migration of Triple‐Negative Breast Cancer Cells

open access: yesAdvanced Science, EarlyView.
Integrated single‐cell mass spectrometry reveals distinct lipidomic signatures that differentiate leader and follower phenotypes in migratory breast cancer cells. Phenotype‐specific alterations across various lipid classes, including fatty acids and phosphatidylcholines, underpin functional heterogeneity.

Autophagy-related molecular clusters identified as indicators for distinguishing active and latent TB infection in pediatric patients

open access: yesBMC Pediatrics
Background Autophagy is crucial for controlling the manifestation of tuberculosis. This study intends to discover autophagy-related molecular clusters as biomarkers for discriminating between latent tuberculosis (LTBI) and active tuberculosis (ATB) in ...
