Dysregulation of SREBP2 induces BACE1 expression
β-Amyloid hyperproduction has been observed in response to alterations in neuronal intracellular cholesterol storage, efflux, and synthesis, induced in rats by a high-fat diet. It has been suggested that cholesterol homeostasis is altered in Alzheimer's disease resulting in higher β- and γ-secretase activity.

Abstract Large clinical data underscore that heart failure is independently associated to an increased risk of negative cognitive outcome and dementia. Emerging evidence suggests that cerebral hypoperfusion, stemming from reduced cardiac output and vascular pathology, may contribute to the largely overlapping vascular dementia and Alzheimer's disease ...

Destruxin E Decreases Beta-Amyloid Generation by Reducing Colocalization of Beta-Amyloid-Cleaving Enzyme 1 and Beta-Amyloid Protein Precursor [PDF]
Alzheimer-disease-associated beta-amyloid (A beta) is produced by sequential endoproteolysis of beta-amyloid protein precursor (beta APP): the extracellular portion is shed by cleavage in the juxtamembrane region by beta-amyloid-cleaving enzyme (BACE ...

SUMOylation, a dynamic post‐translational modification, acts as a master regulator at the heart of tumor malignancy. Our work delineates how the SUMOylation cycle—mediated by E1/E2/E3 enzymes and reversed by SENPs—orchestrates multiple hallmarks of cancer. The central pathway converges on three critical pathological axes: 1.

BACE1 Regulates Hippocampal Astrogenesis via the Jagged1-Notch Pathway
BACE1 is the sole secretase for generating β-amyloid (Aβ) in vivo and is being actively pursued as a drug target for the treatment of Alzheimer’s disease.

ABSTRACT Alzheimer's disease (AD) is a debilitating neurodegenerative condition characterized by progressive cognitive impairment, memory deterioration, and neuronal dysfunction. Its complex pathophysiology involves multiple interlinked processes, including amyloid‐β (Aβ) aggregation, tau hyperphosphorylation, oxidative stress, neuroinflammation ...

Abstract INTRODUCTION Circular RNAs (circRNAs) are emerging as key regulators of gene expression, synaptic plasticity, and neuronal function in Alzheimer's disease (AD). Here, we characterize the biological actions of circPDE4B, a highly expressed circRNA markedly reduced in AD. METHODS circPDE4B knockdown in neuronal progenitor cells was combined with

Asynchronous Evolutionary Origins of A and BACE1 [PDF]
Neurodegenerative plaques characteristic of Alzheimer's disease (AD) are composed of amyloid beta (Aβ) peptide, which is proteolyzed from amyloid precursor protein (APP) by β-secretase (beta-site APP cleaving enzyme [BACE1]) and γ-secretase. Although γ-secretase has essential functions across metazoans, no essential roles have been identified for BACE1

This study reveals that Urolithin A (UA) counteracts alcohol‐induced cognitive and social dysfunction (AICSD) via a gut microbiome‐dependent mechanism. UA‐enriched Bacteroids sartorii and Parabacteroids distasonis elevate anandamide (AEA), which activates the CB1R‐DRD2‐Rap1 signaling cascade to drive synaptic repair and reduce neuroinflammation ...

β‐Secretase (BACE1) is a major drug target for combating Alzheimer's disease (AD). Here we show that BACE1−/− mice develop significant retinal pathology including retinal thinning, apoptosis, reduced retinal vascular density and an increase in the age ...
