Nanomaterials for Allergy Diagnosis and Treatment: Advances, Opportunities and Translational Challenges

open access: yesAdvanced Science, EarlyView.
Nanomaterials offer dual applications in allergy management. For diagnosis, nanomaterials enhance analytical sensitivity and improve detection in specific IgE and functional assays such as the basophil activation test (BAT). For allergen‐specific immunotherapy, nanomaterials enable allergen masking and controlled release, and effectively modulate the ...

Targeting p21‐High Senescent Kupffer Cells Nanotherapeutically Potentiates Antitumor Immunity in Advanced Hepatocellular Carcinoma with Portal Vein Tumor Thrombus

open access: yesAdvanced Science, EarlyView.
Schematic illustration of a biomimetic SKEV@AAV platform targeting p21‐high senescent Kupffer cells (sKCs) in hepatocellular carcinoma (HCC) with portal vein tumor thrombus (PVTT). p21‐high sKCs accumulate in the PVTT microenvironment and establish an immunosuppressive, SASP‐rich environment that facilitates tumor cell proliferation and immune evasion.

Altering VP1 and VP2 expression in trans affects the transduction efficiency of AAV9

open access: yesFrontiers in Bioengineering and Biotechnology
Currently, adeno-associated virus (AAV) is one of the most reliable carrier for gene delivery in both proliferating and non-proliferating cells. Stable and long-lasting transgene expression has made this viral vector a key platform for the development of

HMGCR‐Driven Cholesterol Metabolism Promotes Osteoarthritis Progression by Accelerating Synovial Fibroblast Senescence

open access: yesAdvanced Science, EarlyView.
In the pathological context of osteoarthritis (OA), the phosphorylation of AKT1 at Ser473 enhances its binding to Lys140 of Insig1, which facilitates the formation of AKT1–Insig1 complex. Subsequently, the activation of AKT1 promotes the phosphorylation of Insig1 at Ser189, potentially enhancing the dissociation of Insig1 from sterol regulatory element‑

Oligonucleotide conjugated multi-functional adeno-associated viruses

open access: yesScientific Reports, 2018
Recombinant adeno-associated viruses (AAVs) are among the most commonly used vehicles for in vivo gene delivery. However, their tropism is limited, and additionally their efficacy can be negatively affected by prevalence of neutralizing antibodies in ...

SOX5 Orchestrates Malignant Evolution via Promoter‐Centric Chromatin Remodeling in MYC‐Driven B‐Cell Lymphoma

open access: yesAdvanced Science, EarlyView.
In MYC‐enforced B‐cell lymphoma, SOX5 occupies promoter‐proximal regulatory regions and is associated with reduced chromatin accessibility at the PCNP locus. PCNP repression promotes proliferative remodeling by limiting apoptosis and cell‐cycle restraint.

Structure-based engineering of papillomavirus major capsid L1: controlling particle assembly

open access: yesVirology Journal, 2007
The outer shell of the papillomavirus particle is comprised of 72 pentamers of the major capsid L1 protein arranged on a T = 7 icosahedral lattice. The recombinant L1 can form T = 7 virus-like particles in vitro.

Rational Engineering of Recombinant Picornavirus Capsids to Produce Safe, Protective Vaccine Antigen

open access: yesPLoS Pathogens, 2013
Foot-and-mouth disease remains a major plague of livestock and outbreaks are often economically catastrophic. Current inactivated virus vaccines require expensive high containment facilities for their production and maintenance of a cold-chain for their activity. We have addressed both of these major drawbacks.

Targeted Degradation of Picornaviral 3C Protease via PROTACs Confers High Barrier to Viral Resistance and Broad‐Spectrum Antiviral Activity

open access: yesAdvanced Science, EarlyView.
This study reports D34 as the first PROTAC degrader that targets the 3C protease of picornaviruses. D34 effectively degrades EV71 3C protease via the ubiquitin‐proteasome pathway. More importantly, D34 exhibits a high resistance barrier and shows broad‐spectrum antiviral activity against multiple picornaviruses, highlighting its potential as a novel ...

AAV9-Retro mediates efficient transduction with axon terminal absorption and blood–brain barrier transportation

open access: yesMolecular Brain, 2020
Recombinant adeno-associated viruses (rAAVs), particularly those that permit efficient gene transfer to neurons from axonal terminals or across the blood–brain barrier, are useful vehicles for structural and functional studies of the neural circuit and ...
