Construction of cDNA Libraries in Vaccinia Virus

2004
Poxvirus expression vectors have gained widespread use for expression of foreign proteins and as delivery vehicles for vaccine antigens. We have developed a novel method using the poxvirus as a library vector for functional selection of specific cDNA. Poxviruses have several unique and useful properties as a library vector.

cDNA Library Screening

1999
cDNA libraries are collections of recombinant DNA sequences, derived from and representing entire mRNA populations present at a given stage of development or differentiation in certain tissues or cell types. cDNA libraries can be self-made and tailored according to individual experimental needs.

Construction of cDNA Libraries in λgt10 or λgt11

2003
To obtain a cDNA clone of an mRNA, the mRNA must be copied faithfully into DNA, and the cDNA library must be large enough to represent the abundance class that contains the mRNA of interest. For example, in tobacco, Goldberg (1) has shown that it is possible to divide the mRNA population into three classes with most of the mRNAs (11,300) being in the ...

Expression Screening of cDNA Libraries in pEX

2003
Most methods for screening libraries of cDNA clones rely on the selective binding of either nucleic acid probes to cDNA molecules or antibodies to the polypeptide gene product encoded by the cDNA. Although screening of cDNA libraries with synthetic oligonucleotide probes is undoubtedly a powerful procedure, it is not always the easiest way to clone a ...
