Improving selectivity within therapeutic design is critical to ensure targeted delivery to brain tumors. Targeting the cellular membrane composition distinctive of the blood–brain barrier and brain cancer cells is explored in this review. Nanomedicine can be utilized for precise tumor recognition, and detailed mechanistic interactions of therapeutics ...

Sphingosine kinase 1 and 2 regulate the capacity of mesangial cells to resist apoptotic stimuli in an opposing manner [PDF]
Sphingosine kinases (SKs) are key enzymes regulating the production of sphingosine-1-phosphate (S1P), which determines important cell responses including cell growth and death. Here we show that renal mesangial cells isolated from wild-type, SK-1-/-, and

Ceramide in the Molecular Mechanisms of Neuronal Cell Death. The Role of Sphingosine-1-Phosphate [PDF]
Ceramide and sphingosine-1-phosphate (S1P), two important bioactive sphingolipids, have been suggested as being key players in the pathology of Alzheimer’s disease in inflammation and cancer.

Synthesis, molecular modelling and CYP24A1 inhibitory activity of novel of (E)-N-(2-(1H-imidazol-1-yl)-2-(phenylethyl)-3/4-styrylbenzamides [PDF]
CYP24A1 (25-hydroxyvitamin D-24-hydroxylase) is a useful enzyme target for a range of medical conditions including cancer, cardiovascular and autoimmune disease, which show elevated CYP24A1 levels and corresponding reduction of calcitriol (the ...

Plasma ceramide levels are altered in low and normal birth weight men in response to short-term high-fat overfeeding [PDF]
Low birth weight (LBW) individuals have an increased risk of developing insulin resistance and type 2 diabetes compared with normal birth weight (NBW) individuals. We hypothesised that LBW individuals exhibit an increased fatty acid flux into lipogenesis

A Somatic Mutation in HIF‐2α Affects Glucose and Lipid Metabolism via CD36 and Ceramide
A mutation within the C‐terminal transactivation domain of HIF‐2α enhances Cd36 expression by increased transactivation activity, leading to ceramide accumulation and disruption of glucose and lipid metabolism. Inhibition of ceramide synthesis by myriocin reduces ceramide levels and ameliorates these metabolic disturbances.

Deletion of ASAH2 in intestinal epithelial cells induces the accumulation of ganglioside GD3 via regulating ST8SIA1. GD3 is identified as the glycolipid ligand for the inhibitory receptor Siglec‐E on macrophages. GD3/Siglec‐E ligation polarizes macrophages and promotes the proliferation of colonic ST2+ T regulatory cells via the induction of IL‐33. The
