Bone growth during rapamycin therapy in young rats
Background Rapamycin is an effective immunosuppressant widely used to maintain the renal allograft in pediatric patients. Linear growth may be adversely affected in young children since rapamycin has potent anti-proliferative and anti-angiogenic ...

The TGFβ type I receptor TGFβRI functions as an inhibitor of BMP signaling in cartilage. [PDF]
The type I TGFβ receptor TGFβRI (encoded by Tgfbr1) was ablated in cartilage. The resulting Tgfbr1 Col2 mice exhibited lethal chondrodysplasia. Similar defects were not seen in mice lacking the type II TGFβ receptor or SMADs 2 and 3, the intracellular ...

c-Maf Transcription Factor Regulates ADAMTS-12 Expression in Human Chondrogenic Cells. [PDF]
ObjectiveADAMTS (a disintegrin and metalloproteinase with thrombospondin type-1 motif) zinc metalloproteinases are important during the synthesis and breakdown of cartilage extracellular matrix.

KIF22 regulates mitosis and proliferation of chondrocyte cells
Point mutations in KIF22 have been linked to spondyloepimetaphyseal dysplasia with joint laxity, type 2 (SEMDJL2). Skeletal features of SEMDJL2 include short stature and joint laxity. Mechanisms underlying these limb abnormalities are unknown. Here in this manuscript, we have investigated the function of KIF22 in chondrocytes.

Human periosteum‐derived cell spheroids bioprinted at high density within a hyaluronic acid matrix promote fusion and hypertrophic cartilage formation in vitro. Early encapsulation enhances spheroid interaction and matrix maturation, generating scalable cartilage templates intended for endochondral bone regeneration.

Growth plate closure and therapeutic interventions [PDF]
Height gains result from longitudinal bone growth, which is largely dependent on chondrocyte differentiation and proliferation within the growth plates of long bones. The growth plate, that is, the epiphyseal plate, is divided into resting, proliferative,

Microengineered Gradient Hydrogels for Mechanobiology
Gradient hydrogels are used to mimic the mechanical heterogeneity in native tissues, offering powerful in vitro platforms to study cell‐material interactions in diverse pathophysiological contexts. Here, we present a comprehensive review of the design and experimental considerations for stiffness gradient hydrogels, discussing exemplary achievements ...
