Circular dichroism of C5a anaphylatoxin of porcine complement

Biochemical and Biophysical Research Communications, 1974
Abstract The far-ultraviolet circular dichroism spectrum of C5a anaphylatoxin of porcine complement implies that it has a substantial content of helical structure. The circular dichroism spectra of C5a in the 200–250 nm region at pH 7.2 and 3.7 are nearly identical and resemble those of C3a anaphylatoxin.

C5a, but not C5a‐des Arg, induces upregulation of heteromer formation between complement C5a receptors C5aR and C5L2

Immunology & Cell Biology, 2013
Receptors for C5a have an important role in innate immunity and inflammation where their expression and activation is tightly regulated. There are two known receptors for C5a: the C5a receptor (C5aR) and the C5a receptor like‐2 (C5L2) receptor. Here we hypothesized that activation of C5aR might lead to heteromer formation with C5L2, as a downregulatory

Complement C5A regulates prolabor mediators in human placenta.

Biology of reproduction, 2012
Human preterm and term parturition is associated with inflammatory cascades in the uteroplacental unit. Activation of the complement cascade releases potent proinflammatory mediators, including the anaphylatoxin C5a, which exerts its biological effects through its receptors, C5AR (also known as CD88) and C5L2, official symbol GPR77.

Complement C5a Receptor Assay for High Throughput Screening

Journal of Receptor Research, 1991
The complement C5a receptor on U937 cells, a human histiocytic lymphoma cell line, stimulated with dibutyryl-cAMP have been stabilized for at least 3 months at a dilute, ready to use concentration. [125I]-Bolton Hunter labeled C5a, (recombinant, human) has been prepared by reverse phase HPLC to 2200 Ci/mmol. Using a filtration binding assay the Kd from

The effect of complement C5a on mitochondrial functions of PC12 cells

NeuroReport, 2011
C5a is thought to play a role during complement-activated neuronal apoptotic cell death in the central nervous system. The mechanisms responsible are however not well-understood. As mitochondria play a key role during apoptosis, we investigated mitochondria as a potential target for C5a.

Complement C5a inhibition reduces atherosclerosis in ApoE –/– mice

The FASEB Journal, 2011
ABSTRACT The complement C5a receptor, CD88, is present on many of the cells found within human atherosclerotic plaques, but little is known about the role of C5a in atherogenesis. Using real‐time PCR, we determined that ApoE –/–

Site directed mutagenesis of the complement C5a receptor—Examination of a model for its interaction with the ligand C5a

Molecular Immunology, 1994
C5a is a 74 amino acid peptide cleaved from the fifth component of the complement system after activation of either the alternative or classical pathways. It is a potent chemoattractant for neutrophils and monocytes binding to identical receptors on the cell surface.

Complement in asthma: sensitivity to activation and generation of C3a and C5a via the different complement pathways

Translational Research, 2006
Studies in rodent models suggested that complement may play a critical role in susceptibility to airway hyperresponsiveness (AHR) and as a mediator of bronchial obstruction and inflammation in asthma. Complement may participate in susceptibility to asthma because of an intrinsic abnormality in complement activation and generation of C3a, C5a, or other ...

Mesenteric dysfunction after cardiopulmonary bypass: role of complement C5a

The Annals of Thoracic Surgery, 2000
We investigated the effects of cardiopulmonary bypass (CPB) on ileal homeostasis, and the influence of functional inhibition of complement C5a on CPB-induced mesenteric injury.Pigs were perfused on CPB for 1 hour and then perfused off CPB for an additional 2 hours.

The role of complement component C5a in nociceptive sensitization

2018
<p>The complement system is a principal component of innate immunity. Recent studies have underscored the importance of C5a and other complement components in inflammatory and neuropathic pain, although the underlying mechanisms are largely unknown. In particular, it is unclear how the complement system communicates with nociceptors and which ion
