Leptomycin B alters the subcellular distribution of CRM1 (Exportin 1) [PDF]

open access: yesBiochemical and Biophysical Research Communications, 2017
CRM1 (chromosome maintenance region 1, Exportin 1) binds to nuclear export signals and is required for nucleocytoplasmic transport of a large variety of proteins and RNP complexes. Leptomycin B (LMB), the first specific inhibitor of CRM1 identified, binds covalently to cysteine 528 in the nuclear export signal binding region of CRM1 leading to the ...

ERK1/2 phosphorylates HIF-2α and regulates its activity by controlling its CRM1-dependent nuclear shuttling

open access: yesJournal of Cell Science, 2019
Hypoxia-inducible factor 2 (HIF-2) is a principal component of the cellular response to oxygen deprivation (hypoxia). Its inducible subunit, HIF-2α (also known as EPAS1), is controlled by oxygen-dependent as well as oxygen-independent mechanisms, such as

Dualistic Roles of High Mobility Group Box 1 in Cancer and Inflammation

open access: yesCancer Medicine, Volume 14, Issue 23, December 2025.
ABSTRACT Background The High Mobility Group Box 1 (HMGB1) protein, a member of the HMG family, plays a crucial role in both cancer progression and inflammatory responses. HMGB1 can act as a damage‐associated molecular pattern (DAMP) to activate immune responses and modulate inflammation.

370. CRM1 Enhances the Lentiviral Vector Production

open access: yesMolecular Therapy, 2006
Lentiviral vectors derived from human immunodeficiency virus 1 (HIV-1) are increasingly used in the delivery of transgenes into mammalian cells for gene function studies and for potential treatment of human diseases. One of the major barriers of applying HIV vector in disease treatment is to provide sufficient amounts of infectious vectors at high ...

A Mechanosensitive Channel Governs Lipid Flippase-Mediated Echinocandin Resistance in Cryptococcus neoformans

open access: yesmBio, 2019
Echinocandins show fungicidal activity against common invasive mycoses but are ineffective against cryptococcosis. The underlying mechanism for echinocandin resistance in Cryptococcus neoformans remains poorly understood but has been shown to involve ...

Evolutionarily Conserved Sequence Features Regulate the Formation of the FG Network at the Center of the Nuclear Pore Complex. [PDF]

open access: yes, 2015
The nuclear pore complex (NPC) is the portal for bidirectional transportation of cargos between the nucleus and the cytoplasm. While most of the structural elements of the NPC, i.e.

Protein O‐GlcNAcylation in Health and Diseases

open access: yesMedComm, Volume 6, Issue 12, December 2025.
We first introduced the beneficial effects of normal protein O‐GlcNAcylation on health, impacting biological processes such as gene expression, cell cycle progression, metabolism, and signal transduction. Next, we described the role of abnormal protein O‐GlcNAcylation in the development and progression of diseases, including neurodegenerative diseases,

Lactylation of HMGB1 at K177 Drives Nuclear Export of TIAR to Promote Hypoxia‐Induced Stress Granule Formation

open access: yesAdvanced Science, Volume 12, Issue 41, November 6, 2025.
This study uncovers a novel mechanism in which p300‑catalyzed lactylation of HMGB1 triggers nuclear export of the HMGB1‐TIAR complex, driving TIAR‐dependent SGs formation in the cytoplasm. Mass spectrometry and mutagenesis reveals that K177 lactylation is essential for this export and subsequent SGs formation.

Inhibition of CRM1 activity sensitizes endometrial and ovarian cell lines to TRAIL-induced cell death

open access: yesCell Communication and Signaling, 2018
Background CRM1 enrichment has been shown to be indicative of invasive as well as chemoresistant tumors. On the other hand, TRAIL, a powerful and specific anti-tumoral agent, has yet to be used effectively to treat gynecological tumors in patients.

Multi-state recognition pathway of the intrinsically disordered protein kinase inhibitor by protein kinase A

open access: yeseLife, 2020
In the nucleus, the spatiotemporal regulation of the catalytic subunit of cAMP-dependent protein kinase A (PKA-C) is orchestrated by an intrinsically disordered protein kinase inhibitor, PKI, which recruits the CRM1/RanGTP nuclear exporting complex.
