Multiple gene editing in porcine embryos using a combination of microinjection, electroporation, and transfection methods [PDF]

open access: yesVeterinary World, 2022
Background and Aim: Mosaicism – the presence of both wild-type and mutant alleles – is a serious problem for zygotic gene modification through gene editing using the Clustered regularly interspaced short palindromic repeats-Cas9 (CRISPR/ Cas9) system ...

N-Glycolylneuraminic Acid in Animal Models for Human Influenza A Virus

open access: yesViruses, 2021
The first step in influenza virus infection is the binding of hemagglutinin to sialic acid-containing glycans present on the cell surface. Over 50 different sialic acid modifications are known, of which N-acetylneuraminic acid (Neu5Ac) and N ...

N-glycolylneuraminic acid as a carbohydrate cancer biomarker

open access: yesTranslational Oncology, 2023
One of the forms of aberrant glycosylation in human tumors is the expression of N-glycolylneuraminic acid (Neu5Gc). The only known enzyme to biosynthesize Neu5Gc in mammals, cytidine-5′-monophosphate-N-acetylneuraminic acid (CMAH), appears to be ...

Protein engineering of CMP kinases to improve thermal stability and resultant production of 3′-sialyllactose

open access: yesBiotechnology & Biotechnological Equipment, 2022
Protein engineering is an effective method to improve the thermal stability of biological macromolecules. Cytidine monophosphate kinase (CMK) is a key enzyme involved in nucleotide metabolism and participates in the production of cytidine triphosphate ...

Metabolomic Insights Into the Impact of a Maternal Western-Style Diet and Leptin Supplementation During Suckling on Adult Rat Offspring. [PDF]

open access: yesFASEB J
Plasma metabolites in adult offspring reflecting metabolic programming during critical developmental windows. ABSTRACT Perinatal nutrition influences offspring metabolic phenotype and obesity risk. This study examines how a maternal obesogenic diet and offspring's leptin supplementation during suckling affect the metabolome in offspring adulthood ...

Triple gene editing in porcine embryos using electroporation alone or in combination with microinjection [PDF]

open access: yesVeterinary World, 2022
Background and Aim: We previously developed the gene-editing by electroporation (EP) of Cas9 protein method, in which the CRISPR/Cas9 system was introduced into porcine in vitro fertilized (IVF) zygotes through EP to disrupt a target gene.

Tumor desialylation surpasses anti-PD-L1 checkpoint therapy in restoring anti-tumor immunity in a murine model for colorectal cancer. [PDF]

open access: yesInt J Cancer
What's new? Our research highlights how tumor sialylation affects tumor growth and reshapes the immune tumor microenvironment in colorectal cancer. We demonstrate for the first time that complete removal of sialic acids from colorectal tumor cells elicits a robust anti‐tumor immune response, but does not enhance anti‐PD‐L1 blockade efficacy.

Enhanced Bacterial α(2,6)-Sialyltransferase Reaction through an Inhibition of Its Inherent Sialidase Activity by Dephosphorylation of Cytidine-5'-Monophosphate. [PDF]

open access: yesPLoS ONE, 2015
Bacterial α(2,6)-sialyltransferases (STs) from Photobacterium damsela, Photobacterium sp. JT-ISH-224, and P. leiognathi JT-SHIZ-145 were recombinantly expressed in Escherichia coli and their ST activities were compared directly using a galactosylated bi ...

Aberrant N-glycolylneuraminic acid in breast MCF-7 cancer cells and cancer stem cells

open access: yesFrontiers in Molecular Biosciences, 2022
N-Glycolylneuraminic acid (Neu5Gc) is not normally detected in humans because humans lack the hydroxylase enzyme that converts cytidine-5′-monophosphate-N-acetylneuraminic acid (CMP-Neu5Ac) to CMP-Neu5Gc; thus, any Neu5Gc appearing in the human body is ...
