Building Accurate Intracellular Polarity Maps through Multiparametric Microscopy

open access: yesMethods and Protocols, 2020
The precise knowledge of intracellular polarity, a physiological parameter that involves complex and intertwined intracellular mechanisms, may be relevant in the study of important diseases like cancer or Alzheimer’s.

Fluorescence lifetime imaging microscopy and its applications in skin cancer diagnosis [PDF]

open access: yesJournal of Innovative Optical Health Sciences, 2019
Fluorescence lifetime (FLT) of fluorophores is sensitive to the changes in their surrounding microenvironment, and hence it can quantitatively reveal the physiological characterization of the tissue under investigation.

A Review of New High-Throughput Methods Designed for Fluorescence Lifetime Sensing From Cells and Tissues

open access: yesFrontiers in Physics, 2021
Though much of the interest in fluorescence in the past has been on measuring spectral qualities such as wavelength and intensity, there are two other highly useful intrinsic properties of fluorescence: lifetime (or decay) and anisotropy (or polarization)

Two-photon-excited fluorescence (TPEF) and fluorescence lifetime imaging (FLIM) with sub-nanosecond pulses and a high analog bandwidth signal detection

open access: yes, 2017
Two-photon excited fluorescence (TPEF) microscopy and fluorescence lifetime imaging (FLIM) are powerful imaging techniques in bio-molecular science. The need for elaborate light sources for TPEF and speed limitations for FLIM, however, hinder an even ...

Recent Advances and the Potential for Clinical Use of Autofluorescence Detection of Extra-Ophthalmic Tissues [PDF]

open access: yes, 2020
The autofluorescence (AF) characteristics of endogenous fluorophores allow the label-free assessment and visualization of cells and tissues of the human body.

Nuclear pore links Fob1‐dependent rDNA damage relocation to lifespan control

open access: yesFEBS Open Bio, EarlyView.
Damaged rDNA accumulates at a specific perinuclear interface that couples nucleolar escape with nuclear envelope association. Nuclear pores at this site help inhibit Fob1‐induced rDNA instability. This spatial organization of damage handling supports a functional link between nuclear architecture, rDNA stability, and replicative lifespan in yeast.

Optimizing photoactivation of PA‐mCherry for optical pooled CRISPR screens

open access: yesFEBS Open Bio, EarlyView.
Photoactivatable PA‐mCherry finds widespread use to optically tag individual cells. However, confocal 405 nm UV laser‐scanning (normal scan) is much less efficient than widefield UV illumination, limiting the use of PA‐mCherry on confocal instruments. We remedy this limitation by reporting that rapid and repeated confocal scanning with a low‐intensity,

Enhancing STED microscopy via fluorescence lifetime unmixing and filtering in two-species SPLIT-STED

open access: yesMethods in Microscopy
Simultaneous super-resolution imaging of multiple fluorophores remains a major challenge in STimulated Emission Depletion (STED) microscopy due to spectral overlap of STED-compatible fluorophores.

Automatic Segmentation of Fluorescence Lifetime Microscopy Images of Cells Using Multi-Resolution Community Detection

open access: yes, 2013
We have developed an automatic method for segmenting fluorescence lifetime (FLT) imaging microscopy (FLIM) images of cells inspired by a multi-resolution community detection (MCD) based network segmentation method.

Two-photon phosphorescence lifetime imaging of cells and tissues using a long-lived cyclometallated (NpyridylCphenylNpyridyl)-C-boolean AND-N-boolean AND Pt(II) complex [PDF]

open access: yes, 2014
Using a combination of multiphoton excitation, confocal scanning, TCSPC and beam blanking in conjunction with a cyclometallated Npyridyl^Cphenyl^Npyridyl Pt(II) complex (1) with a long luminescence lifetime, we demonstrate lifetime mapping of living ...
