Precision genome engineering [PDF]
Biotechnology Genome editing through CRISPR-Cas systems has the potential to correct genetic mutations that occur in diseased cells, such as cancer cells. However, the ability to selectively activate CRISPR-Cas systems in diseased cells is important to ensure that gene editing only occurs where it is wanted. Zhu et al.

Structural biology of ferritin nanocages
Ferritin is a conserved iron‐storage protein that sequesters iron as a ferric mineral core within a nanocage, protecting cells from oxidative damage and maintaining iron homeostasis. This review discusses ferritin biology, structure, and function, and highlights recent cryo‐EM studies revealing mechanisms of ferritinophagy, cellular iron uptake, and ...

Precise genome engineering in Drosophila using prime editing
Significance Precise genome engineering allows researchers to modify gene function, tag endogenous proteins, or model human disease mutations. Here, we adapt prime editing, a new CRISPR-based technology that uses reverse transcription to write precise ...

Tau acetylation at K331 has limited impact on tau pathology in vivo
We mapped tau post‐translational modifications in humanized MAPT knock‐in mice and in amyloid‐bearing double knock‐in mice. Acetylation within the repeat domain, particularly around K331, showed modest increases under amyloid pathology. To test functional relevance, we generated MAPTK331Q knock‐in mice.

Nanoparticle analysis sheds budding insights into genetic drivers of extracellular vesicle biogenesis [PDF]
Background: Extracellular vesicles (EVs) are important mediators of cell-to-cell communication in healthy and pathological environments. Because EVs are present in a variety of biological fluids and contain molecular signatures of their cell or tissue of
