Design and synthesis of a cocaine-diamide hapten for vaccine development
Tetrahedron Letters, 1996Abstract A cocaine-diamide hapten was designed in an effort to obtain a potent, long-lasting anti-cocaine immune response for the treatment of cocaine abuse. The analogue incorporated an amido linker functionality in place of the carbomethoxy group at C-2 and a benzoylamino replacement of the benzoyloxy group at C-3 of the cocaine framework. Compound

Science of The Total Environment, 2022
Pendimethalin (PND) is one of the most widely used selective herbicides, but it is considered a potential human carcinogen and persistent bioaccumulative toxic chemical. Herein, five haptens with carboxylic groups were synthesized based on rational design and used to immunize mice, respectively.

Steroids as haptens: Optimal design of antigens for the formation of antibodies to steroid hormones
Journal of Steroid Biochemistry, 1974Abstract By judicious choice of the site of attachment of steroid haptens to a protein carrier it is possible to elicit the formation of antibodies with enhanced specificity towards selected parts of the steroid molecule. Coupling the steroid through a position remote from the functional groups responsible for its hormonal specificity preserves these

Hapten Design for Anti-addiction Vaccine Development
2015The key scientific challenge in the emerging field of drugs of abuse vaccines is the delineation of the complex factors that determine the immune response to the target drug following administration with a chosen hapten. A concise summary is presented of all the various elements of hapten design for the four major drugs of abuse, along with comparative

Bivalent Hapten-Bearing Peptides Designed for Iodine-131 Pretargeted Radioimmunotherapy
Bioconjugate Chemistry, 1997Pretargeting with bispecific antibodies has been used successfully for tumor detection and is now considered for radioimmunotherapy. The advantages of bivalent haptens have been demonstrated in this context. A series of bivalent molecules allowing efficient labeling with radioactive iodine has been designed for use with this new technology.

Hapten Design for Antibody‐Catalyzed Decarboxylation and Ring‐Opening Reactions of Benzisoxazoles
Israel Journal of Chemistry, 1996AbstractThree benzisoxazole haptens designed to elicit antibody binding sites with widely differing polarity have been synthesized and used to induce antibodies in mice. Monoclonal antibodies were prepared using hybridoma technology, and screened for catalysis of the ring‐opening isomerization and/or decarboxylation of a series of related ...

The Design and Synthesis of a Hapten for 1192U90, a Potential Atypical Antipsychotic Agent
Synthetic Communications, 1996Abstract The design and synthesis of a hapten for a radioimmunoassay procedure for the potential atypical antipsychotic agent 1192U90 (1) is described. The hapten, a butyric acid derivative of ortho-amino benzamide 1192U90, was prepared in five steps starting from 5-hydroxy-2-nitrobenzaldehyde.

Adulterated yohimbine (YHB) in food poses a risk to public health, making it imperative to develop fast and sensitive detection methods. In this study, computational-chemistry-based prediction was employed to design YHB haptens for generating the high-affinity monoclonal antibody Yohi-4A7, which exhibited an optimal half-inhibitory concentration (IC50)

Using Hapten Design to Discover Therapeutic Monoclonal Antibodies for Treating Methamphetamine Abuse
The Journal of Pharmacology and Experimental Therapeutics, 2007When generating monoclonal antibodies (mAb) against small molecules, the chemical composition and molecular orientation of the drug-like hapten on the antigen is a crucial determinant. This is especially important when attempting to discover therapeutic mAb against the drugs of abuse (+)-methamphetamine [(+)-METH], (+)-amphetamine [(+)-AMP], and the ...

A morphine/heroin vaccine with new hapten design attenuates behavioral effects in rats
Journal of Neurochemistry, 2011J. Neurochem.(2011) 10.1111/j.1471‐4159.2011.07502.xAbstractHeroin use has seriously threatened public heath in many countries, but the existing therapies continue to have many limitations. Recently, immunotherapy has shown efficacy in some clinical studies, including vaccines against nicotine and cocaine, but no opioid vaccines have been introduced in
