Membrane Fusion Induced by Neuronal SNAREs Transits through Hemifusion [PDF]

open access: yesJournal of Biological Chemistry, 2005
Synaptic transmission requires the controlled release of neurotransmitter from synaptic vesicles by membrane fusion with the presynaptic plasma membrane. SNAREs are the core constituents of the protein machinery responsible for synaptic membrane fusion. The mechanism by which SNAREs drive membrane fusion is thought to involve a hemifusion intermediate,

The Mechanism of Calcium-Mediated Hemifusion on the Pathway to Fusion [PDF]

open access: yesBiophysical Journal, 2011
In many synthetic and biological membrane systems the pathway to fusion passes through a hemifused intermediate where only proximal leaflets are fused. Evidence suggests the unfused distal leaflets engage one another in a hemifusion diaphragm (HD) but the nature of this intermediate is poorly understood even in protein-free systems where divalent ...

Synaptic proteins promote calcium-triggered fast transition from point contact to full fusion

open access: yeseLife, 2012
The molecular underpinnings of synaptic vesicle fusion for fast neurotransmitter release are still unclear. Here, we used a single vesicle–vesicle system with reconstituted SNARE and synaptotagmin-1 proteoliposomes to decipher the temporal sequence of ...

The diffuse interface description of fluid lipid membranes captures key features of the hemifusion pathway and lateral stress profile. [PDF]

open access: yesPNAS Nexus
Topological transitions of lipid membranes are ubiquitous in key biological processes for cell life, like neurotransmission, fertilization, morphogenesis, and viral infections.

SNAREs can promote complete fusion and hemifusion as alternative outcomes [PDF]

open access: yesThe Journal of Cell Biology, 2005
Using a cell fusion assay, we show here that in addition to complete fusion SNAREs also promote hemifusion as an alternative outcome. Approximately 65% of events resulted in full fusion, and the remaining 35% in hemifusion; of those, approximately two thirds were permanent and approximately one third were reversible.

Cholesterol availability regulates the transition between HIV-1 hemifusion and fusion.

open access: yes, 2020
HIV-1 Env sequentially interacts with CD4 and CCR5 at the boundaries of ordered lipid domains. The progression toward hemifusion is not cholesterol dependent as cells treated with 2DG had a reduced concentration of cholesterol and were able to progress ...

Complexin inhibits spontaneous release and synchronizes Ca2+-triggered synaptic vesicle fusion by distinct mechanisms

open access: yeseLife, 2014
Previously we showed that fast Ca2+-triggered vesicle fusion with reconstituted neuronal SNAREs and synaptotagmin-1 begins from an initial hemifusion-free membrane point contact, rather than a hemifusion diaphragm, using a single vesicle–vesicle lipid ...

Meta-stability of the hemifusion intermediate induced by glycosylphosphatidylinositol-anchored influenza hemagglutinin [PDF]

open access: yes, 1997
Fusion between influenza virus and target membranes is mediated by the viral glycoprotein hemagglutinin (HA). Replacement of the transmembrane domain of HA with a glycosylphosphatidylinositol (GPI) membrane anchor allows lipid mixing but not the ...

Conformational Dynamics Related to Membrane Fusion Observed in Single Ebola GP Molecules

open access: yesProceedings, 2020
The Ebola virus (EBOV) envelope glycoprotein (GP) is a membrane fusion machine required for virus entry into cells. Following the endocytosis of EBOV, the GP1 domain is cleaved by cellular cathepsins in acidic endosomes, exposing a binding site for the ...

Chikungunya virus requires an intact microtubule network for efficient viral genome delivery.

open access: yesPLoS Neglected Tropical Diseases, 2020
Chikungunya virus (CHIKV) is a re-emerging mosquito-borne alphavirus, which has rapidly spread around the globe thereby causing millions of infections. CHIKV is an enveloped virus belonging to the Togaviridae family and enters its host cell primarily via
