Harnessing Repurposed Drugs to Enhance Temozolomide Efficacy in Glioblastoma
ABSTRACT Background Glioblastoma (GB) is the most aggressive primary malignant brain tumor in adults and remains associated with poor survival despite surgical resection followed by radiotherapy and temozolomide (TMZ) chemotherapy. Intrinsic and acquired resistance to TMZ, including MGMT‐dependent DNA repair and activation of pro‐survival pathways ...

CD155 is identified as a key driver of anti‐PD‐1 resistance in NSCLC. CD155 promotes tumour glycolysis and malignant progression via the PI3K/AKT/HIF‐1α signalling axis. AAV9_shCD155 combined with anti‐PD‐1 markedly inhibits tumour growth and promotes immune infiltration. Abstract Objective Anti‐PD‐1 therapy resistance remains a critical barrier in non‐

ROS‐ATM‐CBP axis‐mediated PARP1 lactylation aggravates doxorubicin‐induced cardiotoxicity
ROS function as signalling molecules to promote CBP‐mediated PARP1 lactylation in an ATM dependent manner, which induces PARP1 PARylation and ultimate cell apoptosis. CBP and SIRT1 as specific lactyltransferase and delactylase of PARP1. Lactylation of PARP1 depends on the LDHA activity and lactate production.

Hypoxia, oncogenic signaling, transcriptional and epigenetic remodeling, and H. pylori–microbiota interactions drive the Warburg effect in gastric cancer. Enhanced glycolysis and lactate export support biosynthesis, remodel the tumor microenvironment, promote stromal activation and immune suppression, and reveal actionable biomarkers and therapeutic ...

This review elucidates how lactylation, a novel histone modification driven by lactate, regulates colorectal cancer pathogenesis. We summarize its roles in tumor progression, metastasis, stemness, immunosuppression, and therapy resistance, and discusses the promising therapeutic strategies of targeting the lactylation pathway.
