We present a pathology‐responsive, activity‐based prodrug strategy that tackles multiple hallmarks of Alzheimer's disease (AD). We demonstrate that polymeric Janus nanorods (JNRs) delay both oligomerization and fibrillation of human islet amyloid polypeptide (hIAPP) through high‐affinity and sequence‐specific interactions. JNRs display a β‐hairpin mimetic ligand (LP2) on one face, enabling multivalent binding.
