CSF hypocretin-1 levels in restless legs syndrome

Neurology, 2003
CSF hypocretin-1 levels at 6 PM did not significantly differ between patients with restless legs syndrome (RLS) and control subjects as measured by direct radioimmunoassay and after acid extraction. The authors did not observe significant differences between early onset and late onset RLS.

Cardiovascular effects of hypocretin-1 in nucleus of the solitary tract

American Journal of Physiology-Heart and Circulatory Physiology, 2003
Experiments were done in male Wistar rats to investigate the effects of microinjection of hypocretin-1 (Hcrt-1) into the nucleus of the solitary tract (NTS) on mean arterial pressure (MAP), heart rate (HR), and the baroreflex. In the first series, the distribution of Hcrt-1-like immunoreactivity (Ir) was mapped within the region of NTS.

Hypocretin-1/orexin-A activates subfornical organ neurons of rats

NeuroReport, 2008
Central injection of hypocretins/orexins in rats induces water intake. As the subfornical organ (SFO) plays an important role in drinking behavior, hypocretins may excite SFO neurons. In this study, effects of hypocretins on SFO neurons were investigated electrophysiologically in slice preparations. In extracellular recordings, hypocretin-1 excited SFO

Reduced CSF hypocretin-1 levels are associated with cluster headache

Cephalalgia, 2014
Background Cluster headache (CH) is a debilitating disorder characterized by unilateral, severe pain attacks with accompanying autonomic symptoms, often waking the patient from sleep. As it exhibits strong chronobiological traits and genetic studies have suggested a link with the hypocretin (HCRT) system, the ...

Effects of intranasal hypocretin-1 (orexin A) on sleep in narcolepsy with cataplexy

Sleep Medicine, 2011
The neuropeptides hypocretin-1 and -2 (hcrt-1 and -2, also known as orexin A and B) are crucially involved in the regulation of sleep/wake states. On the one hand, the sleep-wake disorder narcolepsy can be caused by an hcrt-1 deficiency. On the other, intracerebral administration of hcrt-1 produces an increase in wakefulness at the expense of REM sleep

Increased CSF hypocretin-1 (orexin-A) in restless legs syndrome

Neurology, 2002
Hypocretin-1 levels were increased in evening CSF samples from subjects with restless legs syndrome, indicating altered hypocretin transmission in this sleep disorder. Increases in CSF hypocretin-1 levels were most striking in patients with early-onset restless legs syndrome.

Effects of hypocretin‐1 in 192‐IgG–saporin‐lesioned rats

European Journal of Neuroscience, 2006
AbstractHypocretin, also known as orexin, is a neuropeptide located in the perifornical region of the lateral hypothalamus; this region projects to all the major arousal centres including the basal forebrain. The basal forebrain contains a mixed population of neurons, some of which are cholinergic.

A monozygotic twin pair discordant for narcolepsy and CSF hypocretin-1

Neurology, 2004
A genetic basis for narcolepsy is evidenced by a strong association with the HLA DQB1*0602 and the occasional familial occurrence.1 A major discovery was recently made through the identification of orexin/hypocretin deficiency.2,3⇓ While a single mutation in the prepro-hypocretin gene was reported in one atypical patient,3 undetectable levels of CSF ...

Cardioacceleratory responses to hypocretin-1 injections into rostral ventromedial medulla

Brain Research, 2003
Intracisternal injections of hypocretin-1 (hcrt-1) have been shown to elicit sympathoexciatory responses. However, the location of central sites that may mediate these cardiovascular effects have not been clearly elucidated. This study was done in male Wistar rats to investigate the effects of microinjections of hcrt-1 into the rostral ventromedial ...

Repeated measures of hypocretin-1 in Danish and Italian patients with narcolepsy and in controls

Sleep Medicine, 2023
The assay currently used worldwide to measure cerebrospinal fluid hypocretin-1 (CSF-hcrt-1) for diagnosing narcolepsy uses a competitive radioimmunoassay with polyclonal anti-hcrt-1 antibodies. This assay detects multiple hypocretin-1 immunoreactive species in the CSF that are all derived from full-length hcrt-1.
