This study identifies TREM2+ macrophages as key contributors to pathological bone formation in ankylosing spondylitis (AS). These macrophages mediate pathological new bone formation via the CREG1‐IGF2R‐PI3K‐AKT pathway and are regulated by IL‐33 through STAT6 phosphorylation.

We report a family with dentinogenesis imperfecta III. The proband and her mother showed rapid attrition and opalescent discoloration of teeth. The primary teeth showed “shell teeth” radiographically. We performed WES and Sanger sequencing on them and revealed a novel variant (c.38C>A: p.A13E) in the signal peptide region of the DSPP gene.

Embryo‐Derived Cathepsin B Promotes Implantation and Decidualization by Activating Pyroptosis
The schematic view shows that embryo‐derived CTSB promotes implantation and decidualization by activating pyroptosis. Blastocyst‐derived CTSB activates NLRP3‐Caspase 1 to cleave GSDMD and cut pro‐IL‐1β/pro‐IL18 to form mature IL‐1β/IL‐18. The cleaved GSDMD‐N perforates the plasma membrane to release mature IL‐1β/IL‐18.
