Inflammation, Immunity, and Cardiovascular Diseases

open access: yesMed Research, Volume 2, Issue 2, Page 343-370, June 2026.
Cardiovascular stress signals (e.g., hemodynamic shear, oxidized lipids, and ischemia) act on endothelial and immune cells to activate and amplify inflammation through NF‐κB, the NLRP3 inflammasome, and JAK/STAT signaling, inducing proinflammatory cytokines/chemokines (IL‐6, IL‐1β, TNF‐α, and CCL2) and self‐amplifying circuits; clinically, inflammatory

Connexin Regulation and Modulation of Neural Stem Cell Differentiation Induced by Cell‐Permeable Itaconate

open access: yesJournal of Cellular Physiology, Volume 241, Issue 5, May 2026.
The immunometabolite itaconate modulates the connexin profile of neural stem cells, and it upregulates Cx36‐based channels, promoting the differentiation toward a neuronal phenotype. These effects were abolished by connexin blockade, supporting a role for itaconate in the neuroimmune axis in inflammatory and neurodegenerative disorders. ABSTRACT Neural

Deficiency of IRG1/ itaconate aggravates endotoxemia-induced acute lung injury by inhibiting autophagy in mice

open access: yesExperimental Animals, 2023
Itaconate, produced by aconitate decarboxylase 1 (ACOD1), which is encoded by immune-responsive gene 1 (Irg1), is one of the metabolites derived from the tricarboxylic acid cycle. It has been reported that exogenous itaconate plays an anti-inflammatory role in the progression of multiple diseases and pathological processes, including activated ...

/itaconate activates Nrf2/HO-1 pathway and mitigates septic liver injury in mice

open access: yesEuropean Journal of Inflammation
Objectives: Immune-responsive gene 1 ( Irg1 )-catalyzed production of itaconate is a bioactive metabolite with antibacterial, anti-inflammatory and antioxidant properties.

Senescent Factors Suppress Innate Antiviral Immunity in Aged Mice via Two Distinct Mechanisms

open access: yesAging Cell, Volume 25, Issue 4, April 2026.
Four secreted factors from senescent cells, including GDF15, IGF1, IL1α, and IL6, cooperatively suppress antiviral innate immunity. GDF15/IGF1 trigger AKT‐MEK‐mediated inactivation of GSK3β, leading to suppression of the TBK1‐IRF3 axis, whereas IL1α/IL6 induce p52 and RelB to suppress transcription of antiviral genes.

Heterologous expression of Mus musculus immunoresponsive gene 1 (irg1) in Escherichia coli results in itaconate production

open access: yesFrontiers in Microbiology, 2015
Itaconic acid, a C5-dicarboxylic acid, is a potential biobased building block for the polymer industry. It is obtained from the citric acid cycle by decarboxylation of cis-aconitic acid.

LCN2‐ACOD1 Signalling Affects the Post‐Injury Regeneration of Skeletal Muscle Through Mediating Ferroptosis

open access: yesCell Proliferation, Volume 59, Issue 4, April 2026.
LCN2 mediates the suppression of C2C12 myoblast proliferation and myotube formation via erastin‐induced ferroptosis. Mechanistically, the LCN2‐ACOD1 axis regulates skeletal muscle growth via mitochondria‐associated ferroptosis, providing a theoretical basis for the prevention and therapy of muscle‐related diseases.

The NLRP3 Inflammasome: Mechanisms of Activation, Regulation, and Therapeutic Opportunities

open access: yesMedComm, Volume 7, Issue 3, March 2026.
Diverse diseases converge on NLRP3. We depict a discovery‐to‐clinic track: high‐throughput/phenotypic screens, structure‐guided design, and modality innovation (allosteric inhibitors, interface blockers, degraders) deliver third‐generation, disease‐tailored NLRP3 control.

Suppressing neutrophil itaconate production attenuates Mycoplasma pneumoniae pneumonia.

open access: yesPLoS Pathogens
Mycoplasma pneumoniae is a common cause of community-acquired pneumonia in which neutrophils play a critical role. Immune-responsive gene 1 (IRG1), responsible for itaconate production, has emerged as an important regulator of inflammation and infection,
