The individual components of commercial isometamidium do not possess stronger trypanocidal activity than the mixture, nor bypass isometamidium resistance [PDF]
The four components present in the trypanocidal treatment Samorin, the commercially available formulation of isometamidium, were separated and purified by column chromatography. These compounds as well as the Samorin mixture and the other phenanthridine trypanocide, homidium, were tested on Trypanosoma congolense and wild type, diamidine- and ...

Genomic analysis of Isometamidium Chloride resistance in Trypanosoma congolense [PDF]
Isometamidium Chloride (ISM) is one of the principal drugs used to counteract Trypanosoma congolense infection in livestock, both as a prophylactic as well as a curative treatment. However, numerous cases of ISM resistance have been reported in different African regions, representing a significant constraint in the battle against Animal African ...

Identification of a genetic marker for isometamidium chloride resistance in Trypanosoma congolense
Isometamidium chloride has remained a very important prophylactic and therapeutic drug against trypanosomosis in cattle since its introduction into the market in the 1950s with, unfortunately, a concomitant development of resistance in trypanosomosis endemic areas. Amplified Fragment Length Polymorphism (AFLP) was used to compare two isogenic clones of

The relationship between serum concentrations of the prophylactic trypanocidal drug isometamidium chloride and protection against tsetse challenge with two populations of Trypanosoma congolense was investigated in Boran (Bos indicus) cattle, using an isometamidium-ELISA.

Effect of isometamidium hydrochloride on Trypanosoma evansi infections in rats [PDF]
Effect of Isometamidium treatment of rats infected with Trypanosoma evansi was studied under experimental condition. The infection resulted in 100 per cent mortality by 7 day post treatment (DPT) in the infected untreated rats (Gr.II) and Isometamidium-HCl exerted 90 percent anti-trypanosomal efficacy.

Fluorescence analysis of the interaction of isometamidium with Trypanosoma congolense [PDF]
Isometamidium chloride (Samorin) is the only compound recommended for prophylaxis against bovine trypanosomiasis in sub-Saharan Africa. The fluorescence property of this compound was used to investigate the interaction of the molecule with in vitro-derived bloodstream forms of Trypanosoma congolense IL 1180.

In Vitro Uptake of Isometamidium and Diminazene by Trypanosoma brucei [PDF]
The cattle trypanocide, isometamidium, was readily taken up by inactivated Trypanosoma brucei , while its uptake by living parasites was reduced or inhibited by plasma. In both respects isometamidium differs from diminazene.

Generation of Monoclonal Antibodies to the Anti-Trypanosomal Drug Isometamidium
Mice were immunized with either an isometamidium-human serum albumin (HSA) conjugate or an isometamidium-porcine thyroglobulin conjugate (PTG). Thereafter, monoclonal antibodies (MAbs) IL-A 1001, IL-A 1002, IL-A 1003, 5F7.B7, and 5F7.C9 were generated and selected on the basis that they recognized conjugated and unconjugated isometamidium, but lacked ...

Reduced accumulation of isometamidium by drug-resistant Trypanosoma congolense
The accumulation of the trypanocide isometamidium chloride (SamorinĀ®, RMB Animal Health Ltd, UK) by a range of clones of Trypanosoma congolense with varying sensitivity to the drug, was measured by methods based on the fluorescence of isometamidium. Fluorescence microscopy and flow cytometry showed a reduction in drug accumulation by resistant clones ...

Determination of Residual Isometamidium in Cattle Tissues and Milk by HPLC
A determination method for the antitripanosomal agent, isometamidium (IMD) in cattle muscle, liver, fat and milk has been developed by using HPLC. The drug was extracted from a sample with acetonitrile-0.25mol/L ammonium formate methanol solution, and purified by liquid-liquid partition with ethyl acetate and water, followed by extraction of the water ...
