Homoisoflavanone Delays Colorectal Cancer Progression via DNA Damage‐Induced Mitochondrial Apoptosis and Parthanatos‐Like Cell Death

open access: yesAdvanced Science, EarlyView.
Homoisoflavanone (HIF), a bioactive compound isolated from Polygonatum kingianum, selectively suppresses colorectal cancer progression by inducing DNA damage‐mediated mitochondrial apoptosis and parthanatos‐like cell death. HIF triggers mitochondrial dysfunction, including depolarized membrane potential, elevated ROS, and ATP depletion, while impairing

Cryo-EM structures of the BAF-Lamin A/C complex bound to nucleosomes

open access: yesNature Communications
Barrier-to-autointegration factor (BAF) associates with mitotic chromosomes and promotes nuclear envelope assembly by recruiting proteins, such as Lamins, required for the reconstruction of the nuclear envelope and lamina.

Differential involvement of caspase-6 in amyloid-β-induced fragmentation of lamin A and B

open access: yesBiochemistry and Biophysics Reports, 2020
Amyloid-β (Aβ), a peptide implicated in Alzheimer's disease, was shown to cause specific fragmentation of lamin proteins, which was mediated by an unidentified protease named nuclear scaffold protease (NSP) independently of caspase-6.

PDZK1‐ULK1 Axis Triggers Lipophagy to Inhibit Tumor Progression and Sunitinib Resistance in Clear Cell Renal Cell Carcinoma

open access: yesAdvanced Science, EarlyView.
This study reveals that PDZK1 loss impairs ULK1‐mediated lipophagy, driving lipid droplet accumulation and sunitinib resistance in clear cell renal cell carcinoma. PDZK1 inhibits LEF1 nuclear translocation, enhancing ULK1 transcription. Pharmacological ULK1 activation resensitizes resistant tumors to sunitinib.

The Mitochondrial Guardian α‐Amyrin Mitigates Alzheimer's Disease Pathology via Modulation of the DLK‐SARM1‐ULK1 Axis

open access: yesAdvanced Science, EarlyView.
Dietary habits play a key role in chronic diseases, and higher annual consumption of fruit and vegetable may lower risk of dementia. Artificial intelligence predicts the lipid‐like compound α‐Amyrin (αA) from plants with edible peels as a drug candidate against Alzheimer's disease.

The role of LMNA in adipose: a novel mouse model of lipodystrophy based on the Dunnigan-type familial partial lipodystrophy mutation[S]

open access: yesJournal of Lipid Research, 2009
We investigated the role of LMNA in adipose tissue by developing a novel mouse model of lipodystrophy. Transgenic mice were generated that express the LMNA mutation that causes familial partial lipodystrophy of the Dunnigan type (FPLD2).

Lamin A/C Mechanotransduction in Laminopathies

open access: yesCells, 2020
Mechanotransduction translates forces into biological responses and regulates cell functionalities. It is implicated in several diseases, including laminopathies which are pathologies associated with mutations in lamins and lamin-associated proteins.

Material‐Induced Nuclear Deformation Controls Chromatin Architecture in Adipose Stem Cells

open access: yesAdvanced Science, EarlyView.
Tuning cell and cytoskeleton mechanics modulated nuclear shape and heterochromatin organization in ASCs. Distinct cytoskeletal architectures induced nuclear morphologies from oblate to prolate ellipsoids. Large elongated cells with a structured actin cap exhibited high nuclear strain, driving nuclear envelope deformation and heterochromatin ...

A Lamin Family-Based Signature Predicts Prognosis and Immunotherapy Response in Hepatocellular Carcinoma

open access: yesJournal of Immunology Research, 2022
Background. Lamin family members play crucial roles in promoting oncogenesis and cancer development. The values of lamin family in predicting prognosis and immunotherapy response remain largely unclarified.

BGN/MDK Axis in the Melanoma Tumor Microenvironment Strengthens Tumor Malignancy by Modulating Cancer Cells and Cancer‐Associated Fibroblasts Crosstalk

open access: yesAdvanced Science, EarlyView.
This study reveals that m6A regulators cooperatively upregulate BGN in melanoma, promoting malignancy. Within the tumor microenvironment, CAFs show highest BGN expression. The BGN/MDK axis mediates cancer‐stroma crosstalk, driving normal fibroblast (NF) activation and enhancing the pro‐tumor effect of CAFs, highlighting a promising therapeutic target ...
