Engineered Microvascular Model of the Blood–Brain–Tumor Barrier Reveals Endothelial Remodeling in Diffuse Midline Glioma

open access: yesAdvanced Healthcare Materials, EarlyView.
We developed a patient‐derived, functional microfluidic model of the diffuse midline glioma (DMG) blood–brain–tumor barrier (BBTB) comprised of endothelial cells, astrocytes, pericytes, and tumor cells. The system forms perfusable microvasculature, reveals the BBTB retains vascular integrity, identifies DMG‐specific transcriptomic changes distinct from

Efficient hepatocyte differentiation of primary human hepatocyte-derived organoids using three dimensional nanofibers (HYDROX) and their possible application in hepatotoxicity research

open access: yesScientific Reports
Human liver organoids are in vitro three dimensionally (3D) cultured cells that have a bipotent stem cell phenotype. Translational research of human liver organoids for drug discovery has been limited by the challenge of their low hepatic function ...

A Fully Defined GelMA‐Based Matrix Allows Fine Tuning of Tissue‐Relevant Biomechanical and Biochemical Cues for Organoid Culture

open access: yesAdvanced Healthcare Materials, EarlyView.
A fully defined GelMA‐based ECM platform integrates tunable tissue‐relevant stiffness with laminin–entactin biochemical supplementation to support mouse small intestinal organoid growth. Coordinated control of mechanical and biochemical cues promotes robust proliferation, budding morphogenesis, differentiation, and passaging, providing a reproducible ...

Tomographic Printing in a Chip: A Versatile Platform for Biomimetic 3D Organ‐on‐Chip

open access: yesAdvanced Healthcare Materials, EarlyView.
A novel light‐based bioprinting approach, TVAM‐in‐a‐chip, enables rapid, contactless fabrication of biomimetic 3D organ‐on‐chip devices directly within preassembled microfluidic platforms. This strategy supports diverse materials, including cell‐laden resins, and eliminates post‐assembly steps.

Vascularization of Human iPSC‐Derived Kidney Organoids Using Perfusion Culture, Pre‐Vascularized Collagen Scaffolds, and Decellularized Extracellular Matrix

open access: yesAdvanced Healthcare Materials, EarlyView.
An integrated perfusion‐based culture platform combining pre‐vascularized collagen scaffolds, stromal vascular fraction cells, and kidney decellularized extracellular matrix promotes vascularization of human iPSC‐derived kidney organoids. The platform provides evidence of interactions between vascular networks and glomerular‐like structures and enables

Construction of vascularized liver microtissues recapitulates angiocrine-mediated hepatocytes maturation and enhances therapeutic efficacy for acute liver failure

open access: yesBioactive Materials
Liver failure poses a significant challenge for millions of patients. The use of primary human hepatocytes and the engineering of liver organoids or liver tissue provide promising solutions to mitigate the shortage of donor organs.
