The proteasome system in infection: impact of β5 and LMP7 on composition, maturation and quantity of active proteasome complexes. [PDF]

open access: yesPLoS ONE, 2012
Proteasomes are the major enzyme complexes for non-lysosomal protein degradation in eukaryotic cells. Mammals express two sets of catalytic subunits: the constitutive subunits β1, β2 and β5 and the immunosubunits LMP2 (β1i), MECL-1 (β2i) and LMP7 (β5i ...

Immunoproteasome subunit LMP7 Deficiency Improves Obesity and Metabolic Disorders [PDF]

open access: yesScientific Reports, 2015
AbstractInflammation plays an important role in the development of obesity and metabolic disorders; however, it has not been fully understood how inflammation occurs and is regulated in their pathogenesis. Low-molecular mass protein-7 (LMP7) is a proteolytic subunit of the immunoproteasome that shapes the repertoire of antigenic peptides on major ...

Contribution of Antigen-Processing Machinery Genetic Polymorphisms to Atopic Dermatitis

open access: yesLife, 2021
Atopic dermatitis (AD) is a chronic and recurrent inflammatory dermatosis. We recently described an association of the C allele of the single nucleotide polymorphism (SNP) rs26618 in the ERAP1 gene and a synergism of ERAP1 and ERAP2 effects on AD risk ...

Co‐inhibition of immunoproteasome subunits LMP2 and LMP7 is required to block autoimmunity [PDF]

open access: yesThe EMBO Reports, 2018
Cells of hematopoietic origin express high levels of the immunoproteasome, a cytokine-inducible proteasome variant comprising the proteolytic subunits LMP2 (β1i), MECL-1 (β2i), and LMP7 (β5i). Targeting the immunoproteasome in pre-clinical models of autoimmune diseases with the epoxyketone inhibitor ONX 0914 has proven to be effective.

Comparative Assessment of the WNT/β-Catenin Pathway, CacyBP/SIP, and the Immunoproteasome Subunit LMP7 in Various Histological Types of Renal Cell Carcinoma

open access: yesFrontiers in Oncology, 2020
ObjectiveThe Wnt/ß-catenin pathway plays an important role in pathogenesis of variety cancers. Most studies on changes in WNT/β-catenin pathway in renal cell carcinoma (RCC) apply only to clear cell RCC, while there are no comparative assessments of this

Post-transcriptional and epigenetic regulation of antigen processing machinery (APM) components and HLA-I in cervical cancers from Uighur women. [PDF]

open access: yesPLoS ONE, 2012
Normal function of human leukocyte antigen class I (HLA-I) and antigen processing machinery (APM) proteins is required for T cell-mediated anti-tumor or antiviral immunity, whereas the tumor survival indicates a failure of the host in immune surveillance

Inhibition and deficiency of the immunoproteasome subunit LMP7 attenuates LCMV‐induced meningitis [PDF]

open access: yesEuropean Journal of Immunology, 2015
In addition to antigen processing, immunoproteasomes were recently shown to exert functions influencing cytokine production by monocytes and T cells, T‐helper cell differentiation, and T‐cell survival. Moreover, selective inhibition of the immunoproteasome subunit LMP7 ameliorated symptoms of autoimmune diseases including CD4+ T‐cell mediated EAE.

Histone deacetylase‐based dual targeted inhibition in multiple myeloma

open access: yesMedicinal Research Reviews, Volume 43, Issue 6, Page 2177-2236, November 2023., 2023
Abstract Despite enormous advances in terms of therapeutic strategies, multiple myeloma (MM) still remains an incurable disease with MM patients often becoming resistant to standard treatments. To date, multiple combined and targeted therapies have proven to be more beneficial compared to monotherapy approaches, leading to a decrease in drug resistance

High glucose leads to redistribution of the proteasomal system

open access: yesBioFactors, Volume 49, Issue 3, Page 636-645, May/June 2023., 2023
The original confocal microscopic image (RGB, left) was split into its three channels “red,” “green,” and “blue.” The single channels were evaluated separately in an automated process using a modified ImageJ macro. Abstract The impact of high glucose on the cellular redox state, causing both induction of antioxidative systems and also enhanced protein ...

Reduced Expression of the Antigen Processing Machinery Components TAP2, LMP2, and LMP7 in Tonsillar and Base of Tongue Cancer and Implications for Clinical Outcome

open access: yesTranslational Oncology, 2015
OBJECTIVES: Patients with human papillomavirus (HPV)–positive tonsillar squamous cell carcinoma (TSCC) and base of tongue squamous cell carcinoma (BOTSCC) have a better clinical outcome than those with corresponding HPV-negative tumors.
