Drug discovery technologies and strategies for Machupo virus and other New World arenaviruses. [PDF]

open access: yesExpert Opin Drug Discov, 2012
Seven arenaviruses cause viral hemorrhagic fever in humans: the Old World arenaviruses Lassa and Lujo, and the New World Clade B arenaviruses Machupo (MACV), Junín (JUNV), Guanarito (GTOV), Sabiá (SABV), and Chapare (CHPV). All of these viruses are Risk Group 4 biosafety pathogens. MACV causes human disease outbreak with high case-fatality rates.

Family-specific degenerate primer design: a tool to design consensus degenerated oligonucleotides [PDF]

open access: yesBiotechnology Research International, Volume 2013, Issue 1, 2013., 2013
Designing degenerate PCR primers for templates of unknown nucleotide sequence may be a very difficult task. In this paper, we present a new method to design degenerate primers, implemented in family-specific degenerate primer design (FAS-DPD) computer ...

Affinity Maturated Transferrin Receptor Apical Domain Blocks Machupo Virus Glycoprotein Binding

open access: yesJournal of Molecular Biology, 2023
Transferrin receptor 1 (TfR) delivers iron across cellular membranes by shuttling the ion carrier protein transferrin. This ability to deliver large protein ligands inside cells is taken advantage of by pathogens to infiltrate human cells. Notably, the receptor's outermost ectodomain, the apical domain, is used as a point of attachment for several ...

Structural Polypeptides of Machupo Virus

open access: yesJournal of General Virology, 1978
The structural proteins of an arenavirus pathogen, Machupo virus, were compared to the structural proteins of two previously characterized non-pathogenic arenaviruses, Pichinde and Tacaribe, in SDS-polyacrylamide gels. Similarities in mol. wt. of the major structural proteins from both pathogenic and non-pathogenic viruses were apparent; however, some ...

A hTfR1 Receptor-Specific VHH Antibody Neutralizes Pseudoviruses Expressing Glycoproteins from Junín and Machupo Viruses [PDF]

open access: yesViruses
The Junín virus (JUNV) is one of the New World arenaviruses that cause severe hemorrhagic fever. Human transferrin receptor 1 (hTfR1) has been identified as the main receptor for JUNV for virus entry into host cells.

Machupo Virus with Mutations in the Transmembrane Domain and Glycosylation Sites of the Glycoprotein Is Attenuated and Immunogenic in Animal Models of Bolivian Hemorrhagic Fever. [PDF]

open access: yesJ Virol, 2022
For arenaviruses, the only vaccine available is the live attenuated Candid#1 vaccine, a JUNV vaccine approved in Argentina. We and others have found that the glycans on GPC and the F427 residue in the GPC TMD are important for virulence of JUNV. ABSTRACT

A review of emerging health threats from zoonotic New World mammarenaviruses [PDF]

open access: yesBMC Microbiology
Despite repeated spillover transmission and their potential to cause significant morbidity and mortality in human hosts, the New World mammarenaviruses remain largely understudied.

Measurement of Antibodies to Machupo Virus by the Indirect Fluorescent Technique

open access: yesExperimental Biology and Medicine, 1973
Bolivian hemorrhagic fever (BHF) was first recognized in the Province of Beni, Bolivia, in 1959 (1), and cases have continued to occur since then. Recent outbreaks have confirmed the fact that clinical manifestations can be quite nonspecific. Laboratory diagnosis is handicapped by a scarcity of facilities in the endemic region, as well as by the hazard

Structure and stabilization of the antigenic glycoprotein building blocks of the New World mammarenavirus spike complex [PDF]

open access: yesmBio
The spillover of New World (NW) arenaviruses from rodent reservoirs into human populations poses a continued risk to human health. NW arenaviruses present a glycoprotein (GP) complex on the envelope surface of the virion, which orchestrates host cell ...
