AARS1 drives PI3K–AKT–mTOR‐dependent glycolysis in bladder cancer, promoting lactate accumulation, increased lactylation, H3K27la enrichment at the HK2 promoter, and HK2 transcription. HK2 reinforces glycolysis, sustaining a metabolic–epigenetic program associated with tumor progression, recurrence, and metastasis.

Targeting the KAT7/H3K14ac/RAC2 Axis Mediates OXPHOS to Promote Stemness Maintenance and Malignancy in Glioblastoma

SOX2 upregulates KAT7 in glioma stem cells, where KAT7 deposits H3K14 acetylation at the RAC2 promoter to activate RAC2 transcription. RAC2‐GTP subsequently promotes PAK1/2/3 phosphorylation, increasing tricarboxylic acid cycle flux and mitochondrial respiration to support glioblastoma malignancy.

TET1 Inhibition Promotes Therapeutic Sensitivity in TP53-Mutant GBM by Influencing Genome Fragility and Altering TAMs Biology. [PDF]

In TP53mut GBM cells, reduced P53 function is associated with increased TET1 expression. Genetic or pharmacological inhibition of TET1 correlates with genome fragility, including DNA damage, cellular senescence, telomere shortening, and reactive oxygen species accumulation, which may contribute to increased efficacy of antitumor therapy.
