The role of motor proteins in endosomal sorting [PDF]

open access: yes, 2011
Microtubule motor proteins play key roles in the spatial organization of intracellular organelles as well as the transfer of material between them.

Tau acetylation at K331 has limited impact on tau pathology in vivo

open access: yesFEBS Letters, EarlyView.
We mapped tau post‐translational modifications in humanized MAPT knock‐in mice and in amyloid‐bearing double knock‐in mice. Acetylation within the repeat domain, particularly around K331, showed modest increases under amyloid pathology. To test functional relevance, we generated MAPTK331Q knock‐in mice.

Mitochondrial defects in the respiratory complex I contribute to impaired translational initiation via ROS and energy homeostasis in SMA motor neurons

open access: yesActa Neuropathologica Communications, 2020
Spinal muscular atrophy (SMA) is a neuromuscular disease characterized by loss of lower motor neurons, which leads to proximal muscle weakness and atrophy.

The SMN protein is a key regulator of nuclear architecture in differentiating neuroblastoma cells

open access: yes, 2009
The cell nucleus contains two closely related structures, Cajal bodies (CBs) and gems. CBs are the first site of accumulation of newly assembled splicing snRNPs (small nuclear ribonucleoproteins) following their import into the nucleus, before they form ...

Spinal cord trauma and the molecular point of no return [PDF]

open access: yes, 2012
A mechanical trauma to the spinal cord can be followed by the development of irreversible and progressive neurodegeneration, as opposed to a temporary or partially reversible neurological damage.

Calpain small subunit homodimerization is robust and calcium‐independent

open access: yesFEBS Letters, EarlyView.
Calpains dimerize via penta‐EF‐hand (PEF) domains. Using single‐molecule force spectroscopy, we measured the strength and kinetics of PEF–PEF homodimer binding. The interaction is robust, shows a transient conformational step before dissociation, and remains largely insensitive to Ca2+.

Structural insights into an engineered feruloyl esterase with improved MHET degrading properties

open access: yesFEBS Letters, EarlyView.
A feruloyl esterase was engineered to mimic key features of MHETase, enhancing the degradation of PET oligomers. Structural and computational analysis reveal how a point mutation stabilizes the active site and reshapes the binding cleft, expading substrate scope.

The Common Functional Dynamics of Molecular Motor and Switch Proteins [PDF]

open access: yesBiophysical Journal, 2013
Understanding how protein ligand binding can promote distinct conformational states, with different affinities for binding partners, is key to understanding the structural basis of protein efficacy. Here we study eight molecular motor and G-protein families that undergo GTP or ATP associated conformational changes to regulate important cell processes ...
