Physiological and behavioral studies with muscimol

Neurochemical Research, 1980
Muscimol has been used to increase our knowledge of central GABAergic systems, CNS physiology, and behavior. Some studies concerning the neurophysiological and behavioral effects of muscimol and its analogs have been reviewed and analyzed. In vivo iontophoretic studies have greatly increased our knowledge of the active conformation(s) adopted by GABA ...

Effects of muscimol on the crayfish neuromuscular junction

Comparative Biochemistry and Physiology Part C: Comparative Pharmacology, 1981
Abstract 1. The effects of muscimol on the crayfish neuromuscular junction were investigated and compared with those of GABA. 2. When muscimol was ionophoretically applied to the glutamate- and GABA-sensitive spots, a slight depolarization of less than about 1 mV was observed.

A Shorter Synthesis of Muscimol

Synthetic Communications, 1982
Abstract The naturally occurring zwitterionic alkaloid muscimol 3 1 has been of considerable pharmacological interest in recent years, it being the best example of an in vivo active mimetic of the neurotransmitter γ-aminobutyric acid (GABA).2 A number of syntheses of this compound have been reported over the past fourteen years, culminating in the ...

Reaction of Muscimol with 4‐Aminobutyrate Aminotransferase

Journal of Neurochemistry, 1983
Abstract: The reaction of muscimol as amino donor substrate for GABA transaminase (GABA‐T) has been studied using enzyme purified from rabbit brain. Enzyme activity was assayed by measuring the glutamate produced using glutamate dehydrogenase. Kinetic parameters determined at 37°C were for GABA, Km (app) = 1.92 ± 0.24 mM, specific activity = 7.33 ± 0 ...

An Improved Synthesis of Muscimol

Synthetic Communications, 1992
Abstract A regiospecific 1,3-dipolar cycloaddition/elimination is the key-step of a convenient, gram-scale synthesis of the GABA-agonist muscimol (1).

Binding of [3H] muscimol to the retina of rabbit and turtle

Comparative Biochemistry and Physiology Part C: Comparative Pharmacology, 1989
1. We studied the binding of [3H]muscimol to membrane preparations of the retina of rabbit (Lepus cuniculus) and turtle (Pseudemys scripta elegans). 2. In both species, [3H]muscimol binding was maximal at 0 degrees C and decreased with increasing temperature, it was saturable, protein concentration dependent and specific. [3H]muscimol binding in the rat retina

Life Sciences, 1980
Abstract Crude membrane fractions were prepared from rat retinae and used to study the specific binding of [ 3 H]muscimol, a potent GABA agonist. Specific [ 3 H]muscimol binding was enhanced 2–3 fold by pretreatment of the membranes with 0.025% Triton X-100. Two muscimol binding sites were demonstrated with K D values of 4.4 and 12.3 nM.

Improvement in Tardive Dyskinesia After Muscimol Therapy

Archives of General Psychiatry, 1979
Muscimol, thought to be a agonist of gamma-aminobutyric acid (GABA), was administered to eight neuroleptic-free subjects with tardive dyskinesia. At oral dose levels from 5 to 9 mg, involuntary movements were consistently attenuated, usually in the absence of sedation.

Central antihypertensive properties of muscimol and related γ-aminobutyric acid agonists and the interaction of muscimol with baroreceptor reflexes

Canadian Journal of Physiology and Pharmacology, 1979
The central antihypertensive properties of four γ-aminobutyric acid (GABA) analogs were characterized in anesthetized cats with implanted intracerebroventricular cannulae. An intracerebroventricular infusion (icv) of muscimol, 0.1–0.5 μg/min (total dose: 1–5 μg, icv), substantially reduced mean arterial pressure and slightly reduced heart rate.

The Ventromedullary Hypotensive Effect of Muscimol in the Anaesthetized Cat

Clinical and Experimental Hypertension, 1981
Muscimol, a rigid analogue of GABA, was injected into the CNS of urethane-anaesthetized, normotensive cats. Bilateral microinjections of a small dose of muscimol (100 ng/kg, 0.5 microliter on each side) in the nucleus reticularis lateralis (NRL) induced hypotension.
