Altered vascular smooth muscle function in the ApoE knockout mouse during the progression of atherosclerosis [PDF]
Objectives: Relaxation of vascular smooth muscle (VSM) requires re-uptake of cytosolic Ca2+ into the sarcoplasmic reticulum (SR) via the Sarco/Endoplasmic Reticulum Ca2+ ATPase (SERCA), or extrusion via the Plasma Membrane Ca2+ ATPase (PMCA) or sodium ...

Loss of proton‐sensing GPR4 reduces tumor progression in mouse models of colon cancer
G protein‐coupled receptor 4 (GPR4) is a pH‐sensing receptor activated by acidic pH. GPR4 expression is increased in patients with inflammatory bowel disease who are at high risk of developing colorectal cancer. In mouse models, loss of GPR4 attenuated tumor progression. This correlated with increased IL2 and natural killer cell activity.

Ion Channels and Their Regulation in Vascular Smooth Muscle [PDF]
Vascular smooth muscle excitability is exquisitely regulated by different ion channels that control membrane potential (Em) and the magnitude of intracellular calcium inside the cell to induce muscle relaxation or contraction, which significantly ...

Systematic profiling of cancer‐fibroblast interactions reveals drug combinations in ovarian cancer
Fibroblasts, cells in the tumor environment, support ovarian cancer cell growth and alter morphology and drug response. We used fibroblast and cancer cell co‐culture models to test 528 drugs and discovered new drugs for combination treatment. We showed that adding Vorinostat or Birinapant to standard chemotherapy may improve drug response, suggesting ...

A control mechanism for intramural periarterial drainage via astrocytes: How neuronal activity could improve waste clearance from the brain [PDF]
The mechanisms behind waste clearance from deep within the parenchyma of the brain remain unclear to this date. Experimental evidence has shown that one pathway for waste clearance, termed intramural periarterial drainage (IPAD), is the rapid drainage of interstitial fluid (ISF) via basement membranes (BM) of the smooth muscle cells (SMC) of cerebral ...

Segmentation of Anatomical Layers and Artifacts in Intravascular Polarization Sensitive Optical Coherence Tomography Using Attending Physician and Boundary Cardinality Losses [PDF]
Intravascular ultrasound and optical coherence tomography are widely available for characterizing coronary stenoses and provide critical vessel parameters to optimize percutaneous intervention. Intravascular polarization-sensitive optical coherence tomography (PS-OCT) simultaneously provides high-resolution cross-sectional images of vascular structures

Loss of primary cilia in endothelial cells promotes EndMT and vascular abnormalities in the ovarian tumor microenvironment through EphA2 activation. Using human samples, in vitro models, and endothelial‐specific Kif3a‐knockout mice, we show that primary cilia loss drives the acquisition of cancer‐associated fibroblast‐like phenotypes, thereby ...

Uncertainty quantification of a three-dimensional in-stent restenosis model with surrogate modelling [PDF]
In-Stent Restenosis is a recurrence of coronary artery narrowing due to vascular injury caused by balloon dilation and stent placement. It may lead to the relapse of angina symptoms or to an acute coronary syndrome. An uncertainty quantification of a model for In-Stent Restenosis with four uncertain parameters (endothelium regeneration time, the ...

Genomics‐led approach to drug testing in models of undifferentiated pleomorphic sarcoma
GA text Genomic data from undifferentiated pleomorphic sarcoma patients and preclinical models were used to inform a targeted drug screen. Selected compounds were tested in 2D and 3D cultures of UPS cell lines. A combination of trametinib and infigratinib was synergistic in the majority of UPS cell lines tested, which was further confirmed in an ex ...

Serotonylation of vascular proteins important to contraction. [PDF]
BACKGROUND:Serotonin (5-hydroxytryptamine, 5-HT) was named for its source (sero-) and ability to modify smooth muscle tone (tonin). The biological effects of 5-HT are believed to be carried out by stimulation of serotonin receptors at the plasma membrane.
