An engineered Trojan Mycobacterium, developed via a “peeling‐off and masking‐up” strategy, functions as a powerful splenic myelopoiesis converter. This spleen‐targeted nanomedicine induces trained immunity to reprogram immunosuppressive myeloid reservoirs into antitumoral effectors.

Genome‐Wide CRISPR Screen Identifies a microRNA Orchestrating Pleiotropic Resistance to Targeted Therapy and T Cell Immunity in Melanoma

A genome‐wide microRNA CRISPR screen identifies miR‐18a as a master regulator of cross‐resistance in melanoma. Loss of miR‐18a activates the AJUBA–YAP/Hippo axis to confer BRAFi resistance and enhances THBS1–CD47 interaction to impair CD8+ T cell immunity. hnRNP A1 is identified as an upstream regulator of miR‐18a processing.

m6A‐Mediated Glycolysis by IL‐37 Drives T Cell Metabolic Reprogramming to Regulate Colitis

This study identifies an IL‐37/SIGIRR‐METTL14 regulatory axis that suppresses global m6A modification in CD4+ T cells. IL‐37 signaling, mediated through SIGIRR, inhibits IRAK4 and JNK phosphorylation, leading to downregulation of the methyltransferase METTL14.

SummaryMyeloid dendritic cells (MDC) play an important role in antigen‐specific immunity and tolerance. In transplantation setting donor‐derived MDC are a promising tool to realize donor‐specific tolerance.
